Proximity Ligation Assay for Protein Interactions

Check if the same lab product or an alternative is used in the 5 most similar protocols
The Duolink PLA red mouse/rabbit kit was used to perform proximity ligation assay per manufacturer’s protocols. Briefly, cells were plated in 8-well chamber slides overnight. The next day, media was aspirated, cells were fixed with 4% formaldehyde with methanol permeabilization. Cells were then blocked with Duolink blocking solution and incubated with primary antibody overnight. Next, Duolink PLA probes were incubated with the samples followed by ligation and amplification. Finally, the slides were mounted with a coverslip using Duolink in situ mounting medium with DAPI, and the slides were either imaged 15 minutes later or kept at −20°C until imaging using a Leica SP5 Inverted Confocal Microscope (DMI6000CS) with the 405 nm and 647 nm lasers for excitation.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
