Evaluating HCC Drug Sensitivity

Two 3D-HCC cancer in vitro models (CSM and ASM) were used in drug sensitivity assays to identify changes in drug sensitivity for each model. The drugs most commonly used for HCC were selected. Sorafenib (S7397), Cabozantinib (S1119), Lenvatinib (S1164), Regorafenib (S1178), 5-FU (S1209), and DOX (S1208) were purchased from Selleckchem (Houston, TX, USA). Drugs were dissolved in a stock solution of dimethyl sulfoxide (DMSO, 100 mM).

Cytotoxicity Assays and Reagents

CCK-8 assay kit (ALX-850–039) was purchased from Enzo Life Sciences (Farmingdale, NY). 5-fluorouracil (5-FU, F6627), doxorubicin (D1515) and ketone body assay kit (MAK134) were obtained from Sigma-Aldrich (St. Louis, MO). Cisplatin (S1166) and sorafenib (S7397) were from Selleck Chemicals (Houston, TX). The antibodies used in this study included anti-SLC13A5 (ab89181) from Abcam, anti-cleaved Caspase-3 (9661S) from Cell Signaling Technology and anti-β-actin (A1978) from Sigma-Aldrich. Oligonucleotide primers were synthesized by Integrated DNA Technologies (Coralville, IA). Unless otherwise specified, all cell culture reagents were purchased from Sigma-Aldrich or Life Technologies.

Huh7 and Hep3B Cell Line Culture

Huh7 and Hep3B cell lines were purchased from the National Collection of Authenticated Cell Cultures (Shanghai, China) and were cultured in complete medium containing Dulbecco’s modified Eagle’s medium (HyClone, Logan, UT, USA) supplemented with 10% fetal bovine serum (Gibco, Grand Island, NY, USA), 1000 U/mL penicillin, and 100 μg/mL streptomycin (HyClone, Logan, UT, USA), and were grown in a humidified air atmosphere containing 5% CO2 at 37 °C. All cell lines were analyzed by STR profiling for cell line authentication and routine mycoplasma detection. Sorafenib (S7397) was purchased from Selleckchem (Houston, TX, USA).
