Mouse Models for Gut Microbiome Studies

C57BL/6, 7B8Tg, zDC-DTR, CCR7−/−, CCR2−/−, BATF3−/−, CX3CR1gfp/gfp, IL-6−/−, MHCIIfl/fl, STAT3fl/fl, IL-6Rαfl/fl, CD4Cre, LysmCre, CD11cCre mice were purchased from The Jackson Laboratory. Mice were bred and maintained at the specific pathogen free animal facilities of the Medical University of South Carolina and later at the Ohio State University. All experiments were performed under protocols approved by Institutional Animal Care and Use Committee at both Universities. Mouse colonies were screened regularly for the presence of fecal SFB. Mice contaminated with SFB was treated with ampicillin in drinking water (1g/ml) for 2 weeks and let recovered before being used for experiments. SFB donor mice include C57BL/6 mice sourced from Taconic and in house SFB+ mice.

Genetically Engineered Mouse Models for Neuroinflammation

Inbred female C57BL/6J, Rag1-/-, Rag2-/-, IL-6-/- and IL-10-/- mice were obtained from the Jackson Laboratory (Bar Harbor, ME). IL-17A-/-, CD11cdnTGF-RII, and CD4dnTGF-RII mice were described previously (38 (link)–40 (link)). IL-17RC-/- mice were developed by Genentech and were obtained from the Mutant Mouse Regional Resource Center (University of California, Davis). IL-17RA-/- and IL-17RA-/-RC-/- were developed in our Lab and described previously (41 (link)). IL-17RD-/- mice were obtained from the Mutant Mouse Regional Resource Center (University of California, Davis). All mice used in this study have a B6 background and were bred in-house. Only female (6 to 8-wk-old) mice were used in the study as male mice are more resistance to HSV-IL-2-induced CNS demyelination than female mice (21 (link)). This is comparable to observations made in other models for MS.

Murine Immunology Knockout Strains

C57BL/6 (B6) as well as IL-6−/−, IL-15−/−, perforin−/−, IFNγ−/−, Rag1−/− μMT−/−, T-bet−/−, TAP1−/−, athymic nude mice and OT-II Tg mice on a B6 background were purchased from The Jackson laboratory (Bar Harbor, MI). Qa-1b−/− mice (originally provided by Dr. Peter Jensen, University of Utah) and CD1d−/−, Jα18−/− and MR1−/− mice (originally provided by Dr. Mitch Kronenberg, La Jolla Institute for Immunology, La Jolla, CA), all on a B6 background, were bred in our own facility. PLZF−/−, PLZF-eGFP (PEG) reporter mice and PCre x R26T mice were provided by Dr. Derek Sant’Angelo, Rutgers University, New Jersey. CD8α−/− and CD8β−/− mice were kindly provided by Dr. Dan Littman (NYU). CD25−/−, CD122−/−and IL-7−/− mice were provided by late Dr. Charlie Surh (TSRI). Mice were housed in individually ventilated cages and used at 6–12 wks of age.

Mouse Strains for Immunological Studies

C57BL/6J, KitW-sh/W-sh, Cpa3-Cre, and Il6–/–, Basoph8 (Mcpt8tm1(cre)Lky/J), and iDTR (Gt(ROSA)26Sortm1(HBEGF)Awai/J) mice were obtained from the Jackson Laboratory (Bar Harbor, ME). St2–/–mice were previously described [60 (link)] and backcrossed 8 generations onto the C57BL/6J strain. Il33–/–mice (C57BL/6J background) were provided by Amgen Inc. Mice were maintained in specific pathogen-free conditions at Northwestern University Center for Comparative Medicine. All protocols were approved by the Northwestern University Animal Care and Use Committee. Isoflurane was used for anesthesia and euthanasia, as appropriate. Bone marrow from Cxcl1–/–mice was kindly provided by Dr. Samithamby Jeyaseelan at Louisiana State University (Baton Rouge, LA) (C57BL/6J background) and were previously described [61 (link)].

Genetic Knockout Mouse Models

All animals were on the C57BL/6 genetic background unless otherwise specified. HuLangerin-Cre I-Aβ (10 (link)), huLangerin-Cre MyD88(6 (link)), huLangerin-DTA (11 (link)), mice have all been previously described. Rag1−/−, IL-6−/−, IL-1β−/−, MyD88/TRIF−/− and B6.129P2-Cxcr6tm1Litt were purchased from Jackson Labs (Bar Harbor, ME). IL-23−/− mice were a kind gift from Nico Ghilardi (Genentech, San Francisco CA). NLRP3−/− and ASC−/− mice were acquired from Millenium Pharmacerticals(12 (link)). All experiments were performed with 8–10 week old age- and sex-matched littermates. Mice were housed in specific pathogen free microisolator cages and fed irradiated food and acidified water. The University of Minnesota institutional animal care and use committee approved all animal protocols.

Adenoviral Cytokine Delivery in Mice

Female BALB/c wild-type (6–8 weeks old) were purchased from Charles River Laboratories (Wilmington, MA, USA). Female C57Bl/6 wild-type (6–10 weeks old), IL-6–/– (C57Bl/6 background; 6–8 weeks old), and RELMα–/– mice (C57Bl/6 background; 8–10 weeks old) purchased from The Jackson Laboratory (Bar Harbor, ME, USA) were acclimatized one week prior to experimental procedures, and housed under specific pathogen-free conditions within the McMaster University Central Animal Facility. All experimental procedures were approved by the McMaster University Animal Research Ethics Board. Mice were endo-tracheally administered with sterile PBS or 5 × 107 PFU of AdDel70, AdOSM, or 3 × 107 PFU of AdIL-6. Animals were culled 2, 7, or 14 days following administration.

Mice were housed in our animal facility in accordance with the Guide for the Care and Use of Experimental Animals, published by the Canadian Council on Animal Care; with the assurance number A5802-01 delivered by the Office of Laboratory Animal Welfare (National Institutes of Health). Tlr9 -/-, Ccr2 -/-, Nlrp3 -/-and C57BL/10 mice were obtained from the Centro Nacional de Biotecnología, Madrid, España.
