Gene Expression Analysis in Rat Model

Check if the same lab product or an alternative is used in the 5 most similar protocols
Rats in both groups (n = 6) were deeply anesthetized with sodium pentobarbital (200 mg/kg, i.p.) on PODs 7, 14, 21, and 28. Total RNA was extracted in TRIzol reagent (Invitrogen, TX, United States, Cat# 15596018). Subsequently, a High-Capacity cDNA Reverse Transcription Kit (Vazyme, Nanjing, China, Cat# R223) was used to convert RNA into cDNA, while a SYBR-Green Real-Time PCR Kit (Vazyme, Cat# Q311) and a Bio-Rad MiniOption thermocycler (Bio-Rad, CA, United States) were used for detection. Primer sequences are provided in Table 1. The relative expression levels of RNA were evaluated using the 2−ΔΔCt method.

Quantifying Gene Expression via qRT-PCR

Check if the same lab product or an alternative is used in the 5 most similar protocols
The expression of PAR-1, SIRT1, and thrombin was measured by quantitative Real-Time PCR (qRT-PCR). TRIZOL reagent (Invitrogen, Grand Island, NY, USA) was applied to extract total RNA. Subsequently, the High-Capacity cDNA Reverse Transcription kit (Applied Biosystems; Thermo Fisher Scientific, Inc.) was used to convert RNA into cDNA, and SYBR-Green I Real-Time PCR kit (TaKaRa Biotechnology, Dalian, China), with a Bio-Rad MiniOption thermocycler (Bio-Rad Laboratories, Inc., Hercules, CA, USA), was used for detection. Primer sequences were as follows:

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
