Western Blot Analysis of Autophagy Markers

Liver tissues or cultured cells were homogenized as described previously [40 (link)]. Equal amounts of protein were separated on 12 % gels by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and transferred to polyvinyldifluoride membranes. The membranes were incubated overnight at 4°C with primary antibodies: rabbit anti-HO-1(1:1000, Abcam, Cambridge, UK), rabbit anti-LC3B (1:1000, Abcam), rabbit anti-Atg16L1(1:1000, Abgent, San Diego, CA), rabbit anti-calpain2 (1:500, Cell Signaling Technology, Beverly, MA), rabbit anti-Atg7 (1:500, Cell Signaling Technology) and rabbit anti-glyceraldehyde-3-phosphate dehydrogenase (GAPDH) antibody (1:20000; Sigma-Aldrich). Immunoreactive band signals were developed using ECL system (GE Healthcare, Buckinghamshire, UK) and visualized on the Kodak Image Station (Carestream Health Inc, New York, USA). Relative band densities were quantified using the Image J software (NIH, Bethesda, USA).
