Xenograft and Liver Metastasis Models

For the histological examination, a xenograft model was established. Accutase-dissociated cells (1 × 106 cells) suspended in Matrigel (BD Biosciences) were transplanted subcutaneously into the dorsal flanks of 7-week-old nonobese diabetic/severe combined immunodeficiency mice (CLEA, Tokyo, Japan). The mice were sacrificed when the tumors reached a diameter of 10 mm. For the liver metastasis model, cells (1 × 106 cells) suspended in 80 μL Dulbecco's modified phosphate-buffered saline (D-PBS; Wako Pure Chemical Industries) were injected into the spleen, which was surgically resected 15 min later. Liver metastasis was evaluated 4 weeks later. The mice were weighed weekly, and none lost weight.

(Osaka, Japan). Dimethyl sulfoxide (DMSO) and fetal bovine serum (FBS) were purchased from Sigma-Aldrich, Co., Ltd. (St. Louis, MO, USA). B standard solution, penicillin–streptomycin mixed solution, and the trypsin (2.5 g/L)-EDTA (1 mmol/L) solution were purchased from Nacalai Tesque, Inc. (Kyoto, Japan). The Cell Counting Kit-8 (CCK-8) was purchased from Dojindo Molecular Technologies, Inc. (Kumamoto, Japan).
