Silencing HIF-1α and FOXO3a in HTR8/SVneo Cells

Specific siRNAs for HIF-1α (siHIF-1α; 5′-CAATCAAGAAGTTGCATTA-3′) and FOXO3a (siFOXO3a; 5′-GCACAGAGTTGGATGAAGT-3′) were purchased from Guangzhou RiboBio Co., Ltd. Then, 250 µl RNase-free water was used to the synthesize siRNAs until they reached a storage concentration of 20 µM. HTR8/SVneo cells were cultured in 6-well plates (1×105/well). Cells with 60% confluence were replaced with fresh culture serum-free medium 2 h prior to transient transfection. Using a final concentration of 50 nM siHIF-1α, siFOXO3a and negative control (siNC), 5 µl siRNAs were transfected into cells using 5 µl Lipofectamine 3000 reagent (Invitrogen; Thermo Fisher Scientific, Inc.). After 48 h of transfection, extraction of total RNA or protein of cells was assessed to confirm efficiency for subsequent experiments.

Si-RNAs were purchased from RiboBio (Guangzhou, China), including si-FOXO3A and si-UBC12. MG132, MLN4924, and PD98059 were purchased from Selleck Chemicals (Houston, TX, USA). Cycloheximide (CHX) was purchased from Absin (Shanghai, China).

Lung Cancer Cell Line Transfection

The lung cancer cell lines PC9, A549, H1975, H1299 and H358 were obtained from the Type Culture Collection of the Chinese Academy of Sciences (Shanghai, China) and cultured in Dulbecco's modified Eagle's medium (DMEM; HyClone, Thermo Fisher, USA) containing 0.1 mM sodium pyruvate, 10% FBS, 2 mM l-glutamine, 100 IU/ml penicillin, and 100 μg/ml streptomycin at 37 °C in a chamber (5% CO2 atmosphere). Cells (2 × 106/per well) were seeded in 6-well plates and transfected with 4 μg of the pEGFP-N1 (control), pEGFP-FoxO3a or pEGFP-EPS8, 5 μl of si-FoxO3a or si-EPS8 (Ribobio, China) using Lipofectamine™ 2000 (Invitrogen, Carlsbad, CA, USA) in antibiotic-free medium. After 48 h, the transient cells were collected for further experiments.
