Immunochemical Identification of CNS Cell Types

Basic reagents were purchased from Sigma (St Louis, MO), unless indicated otherwise. Bromodeoxyuridine (BrdU; 5-bromo-2′-deoxyuridine) and TIMP-1 from human neutrophil granulocytes were purchased from Calbiochem (San Diego, CA); mitomycin and N6,2′-O-dibutyryladenosine-3′,5′-cyclic monophosphate sodium salt (dbcAMP) were purchased from Sigma; cholera toxin B (CTB) subunit was purchased from List Biological Laboratories (Campbell, CA); and 4′,6-diamidino-2-phenylindole (DAPI) was purchased from Molecular Probes (Eugene, OR). The antibodies used for immunodetection were as follows: monoclonal mouse anti-BrdU (B2531; Sigma) and polyclonal rabbit anti–glial fibrillary acidic protein (GFAP; Z0334; Dako, Carpinteria, CA; for dual labeling with NG2, Iba1, and GFAP); monoclonal rat anti-BrdU (ab6326; Abcam, Cambridge, MA; for dual labeling with O1); polyclonal goat anti-CTB (703; List Biological Laboratories); polyclonal rabbit anti-NG2 (AB5320; EMD Millipore, Billerica, MA); monoclonal mouse anti-O1 (MAB344; Chemicon, Temecula, CA); monoclonal mouse anti-CD11b (MCA618R; Serotec, Raleigh, NC); polyclonal rabbit anti-Iba1 (019-19741; Wako, Richmond, VA); monoclonal mouse rat anti-CD68 (MCA341R; Serotec); polyclonal goat anti–calcitonin gene–related peptide (CGRP; 1720-9007; Biogenesis, Bournemouth, United Kingdom); and polyclonal rabbit anti-S100 (Z0311; Dako).

Comprehensive Protein Assays and Compound Characterization

These assays were performed as described (Yang et al., 2007 (link)) using antibodies against GAPDH, β-actin, and caspase-3 (Cell Signaling Technology Inc., MA, USA); and Na+/K+-ATPase α1 (Abcam Inc. Cambridge, UK). Tylophorine and Reevesiosides A-I were prepared as described (Chang et al., 2013 (link), Yang et al., 2010 (link)). DMSO (D1435, ≧ 99.5%), digoxin (D6003, ≧ 95%, HPLC), digitoxin (D5878, ≧ 92%, HPLC), digitoxigenin (D9404, 99%, TLC), ouabain (O3125, ≧ 95%, HPLC), dihydroouabain (D0670, ≧ 98.5%, TLC), oleandrin (O9640, ≧ 98%, HPLC), N6,2′-O-Dibutyryladenosine 3′,5′-cyclic monophosphate sodium salt (Db cAMP) (D0627, ≧ 96%, HPLC), and aldosterone (A9477, ≧ 95%, HPLC) were purchased from Sigma-Aldrich (St. Louis, MO, USA).

Differentiation of Adipocytes with Hormones

High glucose (25 mM) Dulbecco's modified eagle's medium (DMEM; D5796), cell culture grade water, N6,2′-O-Dibutyryladenosine 3′,5′-cyclic monophosphate sodium salt (dbcAMP), dexamethasone (Dex) and 3,3′,5-Triiodo-l-thyronine sodium salt (T3) were all purchased from Sigma-Aldrich (Poole, UK). Human Recombinant Des(1–3) IGF-I was purchased from SourceBioscience and other cell culture reagents were purchased from Fisher Scientific (Loughborough, UK). Compounds were prepared as follows: dbcAMP was dissolved in cell culture grade water at 100 mM, Des(1–3) IGF-I in 0.1 M acetic acid at 100 μg/ml, T3 in 0.1 M sodium hydroxide at 100 nM and Dex in absolute ethanol at 10 mM. Compounds were filter-sterilised through a 0.2 μm filter and aliquots stored at −20 °C.
