Lycorine hydrochloride (Figure 1A ) was obtained from Sigma-Aldrich, Inc. (St. Louis, MO, USA). The anti-mouse CTLA-4 (clone 9H10) monoclonal antibody was purchased from BioXCell (West Lebanon, NH, USA). Primary antibodies against cyclin D1, cyclin D3, CDK2, and β-actin were purchased from Cell Signaling Technology, Inc. (Danvers, MA, USA). Purified PE rat anti-mouse CD4, FITC rat anti-mouse Foxp3, APC rat anti-mouse CD8a, PE rat anti-mouse CD62L, and FITC rat anti-mouse CD44 were purchased from BD Biosciences, Inc. (San Jose, CA, USA).

The cells were stained with allophycocyanin (APC)‐Cy7‐conjugated anti‐CD45 (BD Pharmingen), phycoerythrin (PE)‐conjugated anti‐CD11b (BD Pharmingen), APC‐conjugated anti‐Ly6G (BD Pharmingen), BV421‐conjugated anti‐lymphocyte antigen 6 complex locus C (Ly6C) (BD Horizon), FITC Rat Anti‐Mouse CD19 (Biolegend), BV421 Rat Anti‐Mouse CD3 (BD Pharmingen), PE Rat Anti‐Mouse CD4 (Biolegend) and APC Rat Anti‐Mouse CD8a (BD Pharmingen). Dead cells and debris were gated out using forward light scatter, side light scatter, and 7‐aminoactinomycin D (BD Biosciences, San Jose, CA). The fluorescence‐activated cell sorting analysis was performed on a FACS Canto II flow cytometer and analyzed using FlowJo software (BD Biosciences).

Peripheral blood mononuclear cells (PBMCs) were cleared of red blood cells using the red blood cells lysis buffer from Alfa Aesar (Thermo Fisher Scientific, UK) according to the manufacturer’s instructions. Cells were fixed and stained using BD Cytofix/Cytoperm Fixation/Permeabilization Solution (BD, UK) and analysed using a BD Fortessa flow cytometer (BD). Flow cytometry data were analyzed using FlowJo software (Tree Star). PE-Cy7 rat anti-mouse CD11b, APC-eFluor780 rat anti-mouse CD11c, PE rat anti-mouse CD14, Pacific blue rat anti-mouse CD3, eFluor650 rat anti-mouse CD4 and APC rat anti-mouse CD8a were from BD Biosciences (UK). Anti-Flavivirus Group Antigen antibody, clone D1-4G2-4-15 was obtained from Merck (UK). The secondary Alexa Fluor 488-conjugated goat anti-mouse IgG (H+L) was from Thermo Fisher Scientific (UK).
