We used streptozotocin (STZ)-injected C57BL/6J mice and Ins2Akita mice (C57BL/6-Ins2Akita/J Stock No: 003548/Akita, Jackson Laboratories) as experimental mouse models of type1 diabetes mellitus (DM), while homozygote db/db (BKS/db−/−, Jackson Laboratories) mice were used as mouse model for type 2 DM that develops spontaneous glucose intolerance and hyperglycemia at 4–8 weeks. Heterozygote mice (db/+), which usually show normal body weight, blood glucose, and plasma insulin, but have increased metabolic efficiency were also used for comparison (34 (link)). For STZ injected mice, 6–8 weeks old mice were injected with STZ (55mg/kg, intra-peritoneal) for 3 consecutive days as previously described (35 (link)). Mice with plasma glucose level exceeding 250 mg/dl were considered diabetic. All experimental procedures were performed in accordance with the established guidelines of Association of Research in Vision and Ophthalmology statement for the Use of Animals in Ophthalmic and Vision Research, and were approved by the Institutional Animal Care and Use Committee (IACUC) of Augusta University. Blood was collected from cardiac puncture in heparinized coated tubes and the plasma was separated by centrifugation at 1,500 rpm for 20 min.

Triethylammonium
bicarbonate buffer (1 M,
pH 8.5) and aniline were obtained from Sigma-Aldrich (St. Louis, MO).
DiART 6-plex reagents and CILAT 2-plex reagents were synthesized in-house.15 ,19 (link) HPLC-grade solvents were purchased from Honeywell Burdick &
Jackson (Muskegon, MI). 4-[3-(Perfluorooctyl)-propyl-1-oxy]benzaldehyde
was obtained from Fluorous Technologies Inc. (Ambridge, PA). Strepavidin
Sepharose high-performance beads were purchased from GE Healthcare
Life Sciences (Pittsburgh, PA). Diabetic (BKS.Cg-m+/+Leprdb/J, BKS-db/db) and control (BKS-db/+) mice were purchased from Jackson Laboratories (Bar
Harbor, ME). Mice were housed in a pathogen-free environment, with
continuous access to food (Purina 5053 chow) and water on a 12 h light–12
h dark schedule, and cared for following the University of Michigan

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
