Quantifying Cellular Reactive Oxygen

The ROS content in the liver tissue or JS1 cells was measured using a DCFH-DA fluorescence probe by a reactive oxygen species detection kit (Beyotime). 10 μl DCFH-DA working solution was mixed with 190 μl liver homogenate supernatants and then incubated 30 min at 37°C in the dark. The fluorescence intensity was detected at an excitation wavelength of 500 nm and at an emission wavelength of 525 nm using a multimode reader (Varioskan Flash, Thermo Fisher Scientific). Results were from triplicate experiments.
