Twofold serial dilutions of amphotericin B (Sigma-Aldrich), voriconazole (Sigma-Aldrich), posaconazole (Toronto Research Chemicals), isavuconazole (Toronto Research Chemicals), and caspofungin (Sigma) were prepared in 96-well flat-bottom plates (100 μl per well). R. arrhizus or A. fumigatus spores were diluted in RPMI plus 2% glucose at a concentration of 1 × 103 to 2 × 104/ml depending on the pathogen and drug (specified in the figure legends). For membrane integrity staining, Cytotox Green dye (Sartorius; final concentration, 250 nM) was added. One hundred microliters of the fungal suspension was added to each well. All CN Bio components (controller, docking station, and drivers) and consumables (LC-12 plates) were acquired from CN Bio Innovations (Cambridge, UK) (Fig. S1 †).
