Leukocytes Survival Assay with Leukocins

Human leukocytes were collected as described previously [36 (link)], then incubated with different concentration of Leukocins (HlgAB, HlgBC, LukSF and LukED) at room temperature (RT) for 1 h. After incubation, cells were washed twice with PBS, then stained for 30 min at RT with PE-conjugated CD11b (Biolegend, 101,207) and PerCP-conjugated CD14 (Biolegend, 325,632). The stained cells were analyzed by flow cytometry (BD FACSCalibur, San Jose, USA), and the percentage of survival cells from each sample was calculated.
