Breast Cancer Cell Line Culture Protocols

The breast cancer cell lines 4T1, MDA-MB-231, BT549 and HEK 293 human kidney cells were obtained from the American Type Culture Collection (ATCC®). 4T1, MDA-MB-231 and HEK 293 cells were cultured in high glucose DMEM (Gibco®) supplemented with 10% FBS (Gibco®). BT549 cells were cultured in RPMI-1640 (Gibco®) supplemented with 10% FBS. All cells were maintained at 37°C in a humidified 5% CO2 atmosphere. The ALK5 inhibitor A8301, HDAC8 inhibitor PCI-34051 and paclitaxel were obtained from MedChemExpress (MCE®). Other HDAC inhibitors mentioned in manuscript were from Selleck.

Endothelial Cell HDAC Modulation by Romidepsin

Primary HAEC were purchased from the American Type Culture Collection (Catalog No. PCS-100-011, Lot no. 63233442, Manassas, VA) and maintained in Endothelial Growth Medium-2 (Lonza) supplemented with 10% FBS (GIBCO). HDAC inhibitor Romidepsin (HY-15149), RGFP966 (HY-13909) and PCI-34051 (HY-15224), HDAC activator ITSA-1 (HY-100508) and DNMT inhibitor Decitabine (5-Aza-2'-deoxycytidine, DCA, HY-A0004) were products of MedChemExpress. Unless otherwise indicated, HAEC up to passage 10 without serum starvation were pretreated with Romidepsin (40 nM) for 1 h and then stimulated with 0.1 ng/mL TNFα (R&D Systems) for 2 h for mRNA measurement, or for 4 h for Western blot and flow cytometry analyses.

Evaluation of Pharmacological Inhibitors

VPA and 4′,6-diamidino-2phenylindole dihydrochloride (DAPI) were obtained from Sigma-Aldrich (St. Louis, MO, USA). BIRB 0796 was obtained from Axon Medchem BV (Groningen, The Netherlands), whereas TCS JNK6o was from Tocris Bioscience (Bristol, UK).

Epigenetic Mechanisms in B7x Regulation

The primary antibody against RFX-1 was obtained from Abcam Technology (Cambridge, MA). The mouse B7x antibody was generated by our lab (Albert Einstein College of Medicine). The HDAC siRNAs and C/EBP-α siRNA was from Life Technologies, USA.
