Evaluating PANC1 Cell Viability

PANC1 cells (Invitrogen) were plated in 96-well plates at the density of 8000 cells/well and cultured overnight. On the next day, their medium was replaced with fresh medium supplemented with PNGase F, or the pH of the culture medium was adjusted with hydrochloric acid to 3.5 (100 μl/well). After incubation, 10 μl of the cell counting reagent CCK-8 (Vazyme) was added to each well and the cells were incubated for 2 h at 37 °C in 5% CO2. After incubation, the absorbance value of each well was measured at 450 nm. Three independent experiments were performed.
