PLTP Activity Assay Protocol

Cells were seeded at a density of 2 to 5 × 105 cells/well in 6-well plates and grown overnight at 37 °C. The following day, cells were washed with PBS and incubated in NP-40 lysis buffer for 20 min at 4 °C. The protein lysates were diluted (1:500–1:5000) and used in enzymatic activity assays following the instructions in the PLTP Activity Assay Kit (Sigma Aldrich, MAK108) in 96-well opaque-bottom plates. The activity levels of the samples were measured using a Gen5 (Biotek) microplate reader.
