Western Blot Detection of Tagged Proteins

Cell lysates were generated, and proteins resolved by SDS-PAGE and transferred to nitrocellulose as described [19 (link)]. V5-tagged proteins were detected with mouse monoclonal anti-V5 antibody (ThermoFisher) at 0.3 μg/ml. HA-tagged proteins were detected with mouse (Covance) or rat (Roche) monoclonal anti-V5 antibodies at 1 μg/ml or 12.5 ng/ml respectively. Vacuolar phosphatase [20 (link)] was detected with anti-VTC at 1:5000. Bound antibodies were revealed with either IRDye 800CW dye-conjugated goat anti-mouse Ig and IRDye680RD dye-conjugated goat anti-rabbit Ig (LI-COR) at 25 ng/ml and data was visualized on a LI-COR Odyssey.

Western Blot Detection of Tagged Proteins

Check if the same lab product or an alternative is used in the 5 most similar protocols
Parasite lysates were generated, and proteins (5×106 cell equivalents) were resolved by SDS-PAGE and transferred to nitrocellulose as described (Jensen et al., 2003 (link)). V5-tagged proteins were detected with mouse monoclonal anti-V5 antibody clone SV5-Pk1 (ThermoFisher) at 0.3 μg/ml. HA-tagged proteins were detected with either mouse clone 16B12 (Biolegend) or rat clone 3F10 (Roche) monoclonal anti-HA antibodies at 1 μg/ml or 12.5 ng/ml respectively. Rabbit polyclonal antibodies to either phosphoglycerate kinase (PGK) (Parker et al., 1995 (link)) or CK2α (Jensen et al., 2007 (link)) were used at 1:10,000 and 1:5000 respectively. Bound antibodies were revealed with either IRDye 680 dye-conjuguated goat anti-rabbit Ig (Li-COR) or IRDye 800CW dye-conjugated goat anti-mouse Ig (Li-COR) at 25 ng/ml and data was visualized on a Li-COR Odyssey.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
