Western Blot Analysis of Protein Expression

Treated BMDM cells were lysed with RIPA buffer (supplied with cOmplete Protease Inhibitor Cocktail, Merck#11697498001), and protein concentration was determined by Bradford assay. Same protein amounts were loaded onto 4–12% NuPAGE Gels and protein was transferred to PVDF membranes by using Trans-Blot® Turbo Transfer System (Biorad#17001917). Membrane was blocked with 5% milk-TBS, and then incubated with primary antibodies overnight at 4°C. Signal was developed with HRP-conjugated secondary antibodies and Amersham ECL Western Blotting Detection Reagent (Cytiva#RPN2106).
