Protein Expression Analysis in Cardiac Tissue

Total protein was extracted from frozen heart tissues using RIPA buffer containing proteinase and phosphatase inhibitors (Roche; Germany). Protein lysates were separated by SDS–PAGE and transferred to PVDF membranes (Millipore; USA). The membranes were incubated with the following primary antibodies: GSK-3b, pS9-GSK-3b, NOX2 and GAPDH (Abcam; USA) overnight at 4 °C and then incubated with goat anti-rabbit IgG (Abcam; USA). The Western blot bands were detected using an ECL kit (Keygene; China). ImageJ (NIH; USA) was used to quantify the specific protein expression levels.
