Pcdna3.1 hygro expression plasmid

Manufactured by Thermo Fisher Scientific

The PcDNA3.1(+)/hygro expression plasmid is a vector designed for recombinant protein expression in mammalian cell lines. It contains elements necessary for high-level expression of the gene of interest, as well as a hygromycin resistance gene for selection of transfected cells.

Cloning strategies for PrP constructs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cloning strategies used to generate the cDNAs encoding WT, Δ105–125 (ΔCR), or EGFP-tagged PrP have been described elsewhere [59 (link),60 (link)]. The EGFP-PrP construct contains a monomerized version of EGFP inserted after codon 34 of mouse PrPC. The identity of all constructs was confirmed by sequencing the entire coding region. All constructs were cloned into the pcDNA3.1(+)/hygro expression plasmid (Invitrogen).

EGFP-PrP Construct Generation

Check if the same lab product or an alternative is used in the 5 most similar protocols
The EGFP-PrP construct contains a monomerized version of EGFP inserted after codon 34 of mouse PrP57 (link). The identity of all constructs was confirmed by sequencing the entire coding region. All constructs were cloned into the pcDNA3.1(+)/hygro expression plasmid (Invitrogen). The Strep-FLAG pcDNA3.1(+)/G418 NEGR-1 and the anti-NEGR-1 primary antibody were kindly provided by Giovanni Piccoli (University of Trento, Italy)58 (link). All plasmids were transfected using Lipofectamine 2000 (Life Technologies), following manufacturer’s instructions.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
