Synthesis of Jeffamine M-1000 Copolymer

All materials were reagent grade and obtained from Aldrich unless otherwise noted. For purification, NIPAAm was dissolved in hexane at 10 wt% at 50°C and recrystallized at room temperature. 2,2-Azobisisobutyronitrile (AIBN) was dissolved in methanol at 5 wt% at room temperature and recrystallized at -20°C. The polymerization solvent was HPLC grade Tetrahydrofuran (THF). Dichloromethane (DCM) was distilled and stored under nitrogen.
Jeffamine ® M-1000 was donated by Huntsman Corporation (The Woodlands, TX, USA).
Jeffamine ® M-1000 is a heterobifunctional random copolymer of ethylene oxide (EO) and propylene oxide (PO) in a 19:3 EO:PO ratio and approximately 1000 Da molecular weight, with one methoxy end group and one primary amine end group.

Functionalized Polymer Synthesis and Characterization

Poly (maleic anhydride-alt-1-octadecene) (PMAO, Average Mn 30,000-50,000), diisopropylethyl amine (DIEA), ethanolamine, tetrahydrofuran (THF) and dioxane were purchased from Sigma Aldrich. Jeffamine M-1000 (J-1000) and Jeffamine M-2070 (J-2000) polyetheramine (J-2000) were kindly offered by Huntsman Corporation (Texas, U.S.A.). Vitamin E acetate (VEA) was purchased form TCI Europe. The solvents were of analytical grade, Dimethylformamide (DMF) was anhydrous.

Synthesis and Characterization of PE/ZrP Nanocomposites

Zirconyl chloride (ZrOCl2·8H2O, 98%, Sigma-Aldrich), phosphoric acid
(85%, EM Science), and all of the solvents are reagent grade and used
as received. Jeffamine M1000 with a molecular weight of 1000 g/mol
was donated by the Huntsman Corporation. 3-Aminopropyl trimethoxysilane
(≥97%) was purchased from Alfa Aesar. Octadecyl trimethoxysilane
(ODMS) (≥90%) was purchased from Gelest. MPE and PE used in
this study are listed in Table 1. PE1 has a more uniform structure with narrower molecular
weight distribution compared to PE2. As a result, PE1 is a simpler
matrix to study the PE/ZrP interfacial characteristics and their mechanical
property relationship.

Stimuli-Responsive Polymeric Delivery

Materials were reagent grade and obtained from Sigma-Aldrich unless otherwise noted. N-isopropylacrylamide (NIPAAm) was recrystallized from hexane. Azobisisobutyronitrile (AIBN) was recrystallized from methanol. HPLC grade tetrahydrofuran (THF) was used for polymerizations and as the mobile phase for molecular weight determination. Jeffamine® M-1000 was obtained from Huntsman Corporation (The Woodlands, TX, USA). Jeffamine® M-1000 is a random copolymer of ethylene oxide and propylene oxide units in a 19:3 ratio and approximately 1,000 g/mol molecular weight, with one methoxy end group and one primary amine end group. Tobramycin sulfate USP was obtained from Spectrum Chemical (New Brunswick, NJ). Vancomycin hydrochloride USP was obtained from ChemImpex International (Wood Dale, IL).

Synthesis and Characterization of Functionalized Nanoparticles

Yellow-green carboxylate-modified polystyrene NPs of 100 nm and 20 nm nominal diameter were purchased from Invitrogen (PS-COOH100 and PS-COOH20). PS-COOH100 NPs were pegylated through EDAC chemistry using Jeffamine M1000 (Hunts-man) and purified by centrifugal washings (3 × 30 minutes at 17 000 rcf, 20 °C) (PS-PEG). Bare 50 nm silica NPs were purchased from Kisker (SiO 2 ). Carboxylated Fe 3 O 4 NPs were synthesized following the protocol of Sun et al. 38 and coated by poly(maleic)-alt-1-octadecene (Sigma) according to Lin et al. 39 obtaining NPs of about 50 nm in hydrodynamic diameter. For cell uptake studies, Fe 3 O 4 NPs were fluorescently labelled with BODIPY FLEDA (Lifetechnologies) which was attached to the NP surface by EDAC chemistry. Foetal bovine serum (FBS) was purchased from Fisher. Sucrose, sodium phosphate dibasic, potassium phosphate monobasic, sodium chloride and potassium chloride are from Sigma.
