The activity of caspase-3 was measured using the Caspase 3/7 activity assay kit (AAT Bioquest, Inc., USA) according to the manufacturer’s instruction. Briefly, 5 x 104 cells were plated in each well of 96-well plates and incubated overnight in a 5% CO2 incubator. The cells were treated with rJHP0290 (100 ng/ml) and/or CPT (10 μM), as indicated, for 16 h, followed by the addition of 100 μl/well of caspase 3 assay solution. The plate was incubated at room temperature for 1 h, protected from light. The fluorescence intensity was measured at Ex/Em = 350/450 nm using a POLARstar Omega Instrument (BMG Labtech).
