Genetic Manipulation of Murine Models

Mice were purchased from the Jackson Lab with genotypes including C57BL/6 (JAX # 000664), Thy1-EGFP-M (JAX# 007788), Thy1-YFP-16 (JAX #003709), Ai14 (JAX# 007908), Ai 140 (JAX# 030220), Shh-CreERT2 (JAX# 005623) and Gli1-creERT2 (JAX# 007913). All animal experiments were approved by the Institutional Animal Care and Use Committee of the Chinese Institute for Brain Research.
For tamoxifen treatment, tamoxifen (Sigma-Aldrich, T5648) was dissolved in corn oil (Sigma-Aldrich, C8267) at 20 mg/mL. The solution was kept at –20 °C and delivered via intraperitoneal injection or oral gavage for postnatal treatments.

Transgenic Mouse Strains for Neuroscience

All mice were treated in accordance with guidelines of the Uniformed Services University of the Health Sciences (USUHS) and the National Institutes of Health Guide for the Care and Use of Laboratory Animals. Mice were socially housed as 2–4 mice per cage in 35 cm × 16.5 cm × 18 cm cages that contained enrichment objects and maintained a 12-h daytime light cycle (0600–1800) during which all experimental procedures were performed. The following mouse strains were obtained from the Jackson Laboratory (Bar Harbor, ME, USA): NG2CreER (RRID: IMSR_JAX:008538; B6.Cg-Tg(Cspg4-cre/Esr1*)BAkik/J">B6.Cg-Tg(Cspg4-cre/Esr1*)BAkik/J), ROSAmT/mG (RRID:IMSR_JAX:007676; B6.129(Cg)-Gt(ROSA)26Sortm4(ACTB-tdTomato,-EGFP)Luo/J), and Thy1-YFP-16 (RRID:IMSR_JAX:003709; B6.Cg-Tg(Thy1-YFP)16Jrs/J">B6.Cg-Tg(Thy1-YFP)16Jrs/J). Mouse strains were bred as in-house colonies to generate experimental mice. First-generation offspring from NG2CreER and ROSAmTmG breeders were used as NG2CreER;mTmG mice. The total number of Thy1-YFP-16 mice used was 15 (8 male, 7 female) and the total number of NG2CreER;mTmG mice used was 14 (10 male, 4 female). Male and female mice were combined for analyses after separate analyses showed no sex-based differences. Littermates of Thy1-YFP-16 mice or NG2CreER;mTmG mice were used for quantitative analyses. The number of mice for each condition in each experiment is provided in each figure legend.

All experiments involving mice were approved by the Institutional Animal Care and Use Committee (IACUC) of the Weizmann Institute. Mice were housed in a temperature- and humidity-controlled vivarium on a 12 hr light-dark cycle with free access to food and water.
The following strains were used: Piezo2EGFP-IRES-Cre (The Jackson Laboratory, #027719), Gli1CreERT2 (The Jackson Laboratory, #007913), Rosa26tdTomato (The Jackson Laboratory, #007909), and Thy1-YFP16 (The Jackson Laboratory, #003709).
In all experiments, at least three mice from different litters were used. Mice were genotyped by PCR of genomic DNA from ear clips. Primer sequences and amplicon sizes are listed in Table 1.

Mice Aging and TBI Model Study

At the start of experiments, mice were 8–10 weeks old, which approximately corresponds to young adulthood in humans [26 (link)], a patient population at high risk of TBI [73 (link)]. Transgenic mice expressing YFP under the control of the Thy-1 promoter (Thy1-YFP-16) on a C57BL/6 background were acquired from Jackson Laboratories (Bar Harbor, ME). The original Thy1-YFP-16 mice (IMSR Cat# JAX:003,709, RRID:IMSR_JAX:003,709; B6.Cg-Tg(Thy1-YFP)16Jrs/J) were bred in-house to generate the experimental male (n = 3/group) and female (n = 4/group) mice for Study 1 – Confocal Analysis. As no sex-based differences were detected in this initial study, the experiments of Studies 2 and 3 used only male mice. C57BL/6J (IMSR Cat# JAX:000,664, RRID:IMSR_JAX:000,664) mice were purchased from Jackson Laboratories. Mice were socially housed in 35 cm × 16.5 cm × 18 cm enrichment cages (2–5 mice per cage), exposed to 12:12 h light–dark cycle, at controlled room temperature, and with free access to food and water. All experimental procedures were conducted during the daytime light cycle (0600–1800) after the mice had acclimated for at least 3 days in their home cage.
