SGC-7901 cells were seeded in six-well plates at 2×105 cells per well and transfected with polymer/siRNA as described earlier. Cells were harvested and lysed at 3 days post-transfection using radioimmunoprecipitation-assay lysis buffer (Santa Cruz Biotechnology, Dallas, TX, USA). Then, samples were run through 12% sodium dodecyl sulfate polyacrylamide gels and transferred onto polyvinylidene difluoride (PVDF) membranes. Monoclonal rabbit anti-PD-L1 antibody (Abcam, Cambridge, UK) and polyclonal rabbit anti-β-tubulin antibody (Cell Signaling Technology, Danvers, MA, USA) were used for Western blotting. Protein blots on PVDF membranes were imaged using a chemiluminescence-detection method (Merck Millipore, Billerica, MA, USA).
