Synthesis and Purification of Polymeric Fluorescent Probes

DPAEMA (97%) was obtained from Sigma-Aldrich (St. Louis, MO, USA) and purified using a Sigma-Aldrich inhibitor removal column. MPC was purchased from NOF Corp. (Tokyo, Japan), which was synthesized and purified following a previously reported method [28 (link)]. 4-Cyanopentanoic acid dithiobenzoate (CPD) was synthesized as a chain transfer agent (CTA) according to a reported procedure [29 (link)]. Tris(2-carboxyethyl)phosphine hydrochloride (TCEP, 98%), ethylenediamine (>99.0%), and 4,4′-azobis-(4-cyanovaleric acid) (V-501, 98%) were purchased from Wako Pure Chemical (Osaka, Japan) and used without any purification procedure. Pyrene (Py, 97%) was supplied by Wako Pure Chemical and purified via recrystallization from methanol. Alexa Fluor 555 C2 maleimide (A555) and Alexa Fluor 488 C5 maleimide (A488) were purchased from Thermo Fisher Scientific (Tokyo, Japan) and used as received. Methanol and ethanol were dried over 4Å molecular sieves and purified via distillation. Water was purified using an ion-exchange column.
