Automated DNA Extraction from Maternal and Infant Samples

Check if the same lab product or an alternative is used in the 5 most similar protocols
Genomic DNA was extracted from the mothers’ and infants’ samples as described in Mortensen et al., 2016 (link), using the PowerMag Soil DNA Isolation Kit optimized for epMotion (MO-BIO Laboratories, Inc, Carlsberg, CA) using the epMotion robotic platform model (EpMotion 5075VT, Eppendorf, Hamburg, Germany).

Quantification of TMAO, Carnitine, and Related Metabolites

Check if the same lab product or an alternative is used in the 5 most similar protocols
Semifasted EDTA plasma TMAO, carnitine, choline, betaine, and DMG concentrations were measured using 2 previously described analytical assays (29 (link)). Briefly, sample preparation (i.e. protein precipitation, mixing, and filtration) was conducted on an EpMotion 5075vt robot fitted with a thermal mixer and vacuum manifold (Eppendorf). This was followed by UHPLC/MS-MS, performed on a Vanquish UHPLC+ system, coupled with a TSQ Quantiva triple quadrupole mass spectrometer (Thermo Scientific).

DNA Extraction from Swab Samples

Check if the same lab product or an alternative is used in the 5 most similar protocols
The swab tips were submerged in 1 mL of molecular grade water (W4502, Sigma, UK), and bacteria released by 10s manual agitation. DNA extraction from 200 μl of the water was performed using NucleoSpin® 96 soil kit (740787, Macherey-Nagel, Germany) using an EpMotion 5075vt (Eppendorf, USA), following manufacturer instructions, using lysis buffer SL1 with 150 μl Enhancer SX and eluted in 70 μl elution buffer.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
