Quantitative Analysis of Kratom Alkaloids

Chloroquine, 7-hydroxymitragynine, midazolam, mitragynine, and mitragynine-d3 were purchased from Cayman Chemical (Ann Arbor, MI, USA). The kratom alkaloids speciogynine, mitraciliatine, speciociliatine, paynantheine, and isopaynantheine were isolated and purified from Mitragyna speciosa as detailed previously [7 (link)]; all standards were >95% pure as measured by ultraperformance liquid chromatography (UPLC). Alprazolam and tert-butyl methyl ether (t-BME) were purchased from Sigma-Aldrich (St. Louis, MO, USA). Human liver microsomes (HLMs; H0604, mixed biological sex, pool of 15, lot 1010191) and human intestinal microsomes (HIMs; H0610.I, mixed biological sex, pool of 10, lot 1610314) were purchased from XenoTech, LLC (Kansas City, KS, USA). Acetonitrile and methanol were purchased from Fisher Scientific (Fair Lawn, NJ, USA). Blank human plasma (K2EDTA, mixed biological sex, pooled, lot HMN99236) and whole blood (K2EDTA, mixed biological sex, pooled, lot HMN541526) were purchased from BioIVT (Westbury, NY, USA). All other chemicals and reagents were analytical grade.

Quantitative analysis of ketamine and its metabolites

Actoacetanilide was the product of the Tokyo Chemical Industry (Tokyo, Japan). S-Ketamine was obtained from Cayman Chemical Company (Ann Arbor, MI, USA) as a certified reference material (CRM) solution in methanol (1 mg/mL). Before using this reagent, the methanol solvent was evaporated under a stream of argon gas, and the chemical was redissolved in 0.1 M Hepes buffer pH 7.4 containing 60 mM KCl. The concentration of ketamine in the stock solution was determined from its absorbance at 265 nm using the extinction coefficient of 0.562 mM−1cm−1. (S)-norketamine hydrochloride and midazolam were purchased from Cayman Chemical Company (Ann Arbor, MI, USA). Glucose-6-phosphate dehydrogenase from Leuconostoc mesenteroides, NADP, Glucose-6-phosphate, and midazolam were the products of MilliporeSigma (Burlington, MA, USA). All other reagents were of ACS grade and used without additional purification.

Generating TrkA-KFG Mutant Mice for Behavioral Studies

The generation of TrkA-KFG mutant mice has been described elsewhere [20 (link)]. All tests were conducted on at least 3-month-old, age-matched, male mutants, or C57BL/6CR mice. Male mice were used to exclude the effects of sexual dimorphism and estrous phase on behavior. Animals were kept in the NCI vivarium in colony cages at an ambient temperature of 25 ± 2 °C and 45–55% relative humidity with 12 h light: dark cycle. The mice had free access to standard rodent pellet diet and drinking water. Donepezil (3 mg/kg i.p) was obtained from Sigma-Aldrich (USA). Picrotoxin (0.5 mg/kg i.p) and midazolam (1.25 mg/g i.p) were from Cayman Chemicals (USA). The drug dosage was based on results of pilot experiments and earlier publications [21 (link)–23 (link)]. TMT (2, 3, 5-Trimethyl-3-thiazoline) was obtained from PheroTech, Canada. All procedures were approved by NCI-Frederick ACUC committee and followed the National Institutes of Health Guidelines for animal care and use.
