Apoptosis Detection by Annexin V-FITC/PI Assay

An Annexin V-Fluorescein Isothiocyanate (FITC)/Propidium Iodide (PI) Staining kit (BD Biosciences, Franklin Lakes, NJ, USA) was used to detect apoptotic cells. Cells were harvested 48 h following transfection and resuspended in 1X binding buffer at a concentration of 1×106 cells/ml. Subsequently, cells were stained with 5 µl Annexin V-FITC and 5 µl PI for 15 min at room temperature in the dark. Cells were acquired using a BD Accurri C6 flow cytometer (BD Biosciences) and data was analyzed using FlowJo software version 7.6.2 (Tree Star, Inc., Ashland, OR USA).

Measuring Mitochondrial Membrane Potential

To evaluate the mitochondrial membrane potential (MMP), a lipophilic cationic probe 5,5′,6,6′-tetrachloro-1,1′,3,3′-tetraethylbenzimidazol-carbocyanine iodide (JC-1, 5 mg l−1, Biotime) was added to cell cultures for 30 min at 37 °C and fluorescence was measured by using a fluorescent microscope (Olympus, Tokyo, Japan) or a BD Accurri C6 Flow Cytometer. In all, 10 μM CCCP (carbonyl cyanide 3-chlorophenylhydrazone, Beyotime)-pretreated groups were used as positive control. The experiment was repeated at least three times.
