Murine and Human Osteosarcoma Cell Lines

The murine osteosarcoma MOS-J cell line, called MOS-J/Native in this study, derived from a spontaneous C57BL/6 mouse osteosarcoma, was provided by Prof. L. Shultz [21 ]. Two subclones derived from this cell line were also used in the experiments [6 (link)]. The first clone, MOS-J/PG1, revealed a high proliferation rate in vitro in contrast to the second, MOS-J/A3N. These clones, as well as the MOS-J/Native, were grown in RPMI1640 medium (Lonza, Walkersville, MD, USA) supplemented with 5% fetal bovine serum (FBS; Hyclone, Logan, UT, USA) and a mix of 100 U/mL of penicillin and 100 µg/mL of streptomycin (Lonza). The human osteosarcoma cell lines G292 (clone A141B1), KHOS/NP (R-970-5) named HOS in the manuscript, 143B, MG63, SJSA-1, and SaOS2 were purchased at the American Type Culture Collection (ATCC, Manassas, VA, USA), respectively, under references CRL-1423, CRL-1554, CRL-8303, CRL-1427, CRL-2098, and HTB-85. The CAL-72 cell line was purchased at the DSMZ-German Collection of Microorganisms and Cell Cultures (DSMZ, Leibniz Institute, Braunschweig, Germany) under reference ACC-439. G292, HOS, 143B, MG63, SaOS2, and CAL-72 cell lines were cultured in DMEM (Lonza) and the SJSA-1 cell line in RPMI-1640 (Lonza), both supplemented with 10% FBS.

Colorectal Cancer Cell Line Characterization

Human colorectal cancer cell lines (SW1116 and SW837) and normal human fibroblasts (CRL1554) were obtained from the American Type Culture Collection, ATCC (VA, USA). Leibovitz’s L-15 and EMEM (Eagle Minimum Essential Medium), trypsin, penicillin/streptomycin solution and fetal bovine serum (FBS) were obtained from Mediatech Inc. (Herndon, VA, USA). Primers, Taqman probes and all of the reagents for RT-PCR and real-time quantitative PCR (qPCR) were obtained from Applied Biosystems (Carlsbad, CA). The DNA-prep kit was obtained from Beckman & Coulter (Kendall, FL), and an Annexin V-FITC apoptosis detection kit was obtained from Hoffmann-La Roche Inc. (Nutley, NJ, USA). NFkB (p65) transcription factor assay kit was obtained from Cayman Chemical (Ann Arbor, MI, USA) and nuclear/cytosol fractionation kit was purchased from BioVision Inc. (Milipitas, CA, USA). Organic solvents of high-performance liquid chromatography (HPLC) grade were purchased from Fisher Scientific (Atlanta, GA, USA). Drugs, standard ferulic acid (FA) and other chemicals were obtained from Sigma-Aldrich Chemicals (St Louis, MO, USA).

Comparative Analysis of Hepatic Cell Lines and Natural Product Compounds

The hepatic cancer cell lines, Hep3b and HepG2 and the normal human fibroblast cell line CRL1554 were obtained from the American Type Culture Collection (Manassas, VA, USA). HepG2 and Hep3b cells were grown in Eagle's minimal essential medium (EMEM; Gibco; Thermo Fisher Scientific, Inc., Waltham, MA, USA) containing 10% fetal bovine serum (FBS; Gibco; Thermo Fisher Scientific, Inc.) in a 5% CO2 incubator at 37°C. HepG2 cells, which were originally thought to be a HCC cell line (12 (link),13 (link)), have been misidentified and are considered to be a hepatoblastoma cell line (14 (link)). CRL1554 cells were grown in Dulbecco's modified Eagle's medium (DMEM; Gibco; Thermo Fisher Scientific, Inc.) containing 10% FBS in a CO2 incubator at 37°C. Penicillin/streptomycin (Sigma-Aldrich; Merck KGaA, Darmstadt, Germany), gentamicin (Gibco; Thermo Fisher Scientific, Inc.), L-glutamine (Fluka; Honeywell Labs, Muskegon, MI, USA) and sodium hydrogen carbonate were added to all media during preparation. The NPCs were all obtained from Sigma-Aldrich (Merck KGaA) and included homoharringtonine (HHG), Kmf, Cur, Que, Rsv, hesperetin (Hsp), betulinic acid (BetA), indol-3-carbinol (IC3), coumarin (Cmr), sulanidac (Sul), irinotecan (Irt), lycopene (Lyp), silibinin (Sil) and sinirgin (Snn).

SW1116 and SW837 cells were grown in Leibowitz-15 (L15; 90%; Gibco; Thermo Fisher Scientific, Inc.) containing 10% FBS in a non-CO2 incubator. CRL1554 cells were grown in DMEM (90%; Gibco; Thermo Fisher Scientific, Inc.) containing 10% FBS in a CO2 incubator. Penicillin/streptomycin (Sigma-Aldrich; Merck KGaA), gentamicin (Gibco; Thermo Fisher Scientific, Inc.), L-glutamine (Fluka; Biochemika™), and sodium hydrogen carbonate (BDH; GPR™) were added to all media during preparation. The PPCs were all obtained from Sigma-Aldrich (Merck KGaA), including betulinic acid (BetA), coumarin (Cmr), Cur, hesperetin (Hsp), homoharringtonine (HHG), indol-3-carbinol (IC3), irinotecan (Irt), Kmf, lycopene (Lyp), Que, Rsv, silibinin (Sil), sinigrin (Snn), and sulanidac (Sul). Sora was purchased from MedChemExpress.

Cancer and normal cell culture

Human colorectal cancer cell lines (SW1116 and SW837), human breast cancer cell lines (HTB26, HTB132), and normal human fibroblast cells (CRL1554) were obtained from American Type Culture Collection (ATCC; VA, USA). SW1116, SW837, HTB26 and HTB132 cells were cultured in 90% Leibovitz’s L15 medium supplemented with 10% heat-inactivated fetal bovine serum and grown at 37°C in a non-CO2 incubator. CRL1554 cells were cultured in Eagle minimum essential medium, EMEM (90%) supplemented with 10% heat-inactivated fetal bovine serum and grown at 37°C in the presence of 5% CO2 and 95% ambient air.
