Neurobehavioral Phenotype in MRL/lpr Mice

The 8–10–week-old MRL/lpr (stock # 000485) and MRL/+ (stock # 000486) mice were purchased from The Jackson Laboratories (Bar Harbor, ME, USA). Female mice were used unless otherwise specified. NPSLE manifestations are absent in the congenic MRL/+ strain and more prominent in female than in male MRL/lpr mice (18 (link), 19 (link)), and CP infiltrating T cells were found to be rare or diminished in the non-autoimmune control MRL/+ strain and in age matched male MRL/lpr mice, respectively (see below). Hence, MRL/+ or male MRL/lpr mice were used as controls in some experiments. Mice were housed in the animal facility of Albert Einstein College of Medicine until they were 16–18 weeks of age, at which time the MRL/lpr strain exhibits a profound neurobehavioral phenotype including cognitive deficits and depressive like behavior (20 (link)–22 (link)). All animal studies were performed under protocols approved by the Institutional Animal Care and Use Committee of the Albert Einstein College of Medicine.

Genetic Regulation of Autoimmune Disorders

All animal experiments and methods of euthanasia were approved by the Ralph H. Johnson VAMC Institutional Animal Care and Use Committee (IACUC). Mice were housed and maintained under pathogen-free conditions at the Ralph H. Johnson VAMC Animal Care Facility (Charleston, SC). MRL/lpr Fli1+/+ and Fli1+/− mice (6 (link)) and C57BL/6 Fli1+/+ and Fli1+/− mice (16 (link)) were obtained from matings between Fli1+/+ and Fli1+/− mice within each strain in our colony. Wild-type MRL/lpr and C57BL/6 Fli1+/+ mice obtained from Jackson Laboratories (Bar Harbor, ME) were used for matings every other generation to avoid genetic drift in the colonies. Age-matched animals of both genders were used in experiments.

Decoding Lupus Pathogenesis in Mice

All mice, except for the MRL/MpJ-Faslpr/J, were on the C57BL/6J background. All mice were housed and bred in the SUNY Upstate Medical University Animal Care Facility (Syracuse, NY), in accordance with institutional guidelines for animal welfare. All mice used for experiments were at least 6 weeks old, and both male and female mice were used unless otherwise stated. All studies involving animals were approved by the SUNY Upstate Medical University Institutional Animal Care and Use Committee NYSDOH Unit A073 IACUC number 311.

MRL/lpr Mice Breeding and Care

Diverse Mouse Strains for Comprehensive Research

For this study, adult female mice from the following four standard inbred strains were used: C57BL/6 J (The Jackson Laboratory, Bar Harbor, ME, USA); B6.129S6(Cg)-Spp1tm1Blh/J, also known as OPN−/− (The Jackson Laboratory); B6.129P2-Icos/J also know as ICOS−/− (The Jackson Laboratory), BALB/cOlaHsd, or BALB/c mice (Envigo, Huntingdon, UK); and MRL/MpJ-Faslpr/J, commonly known as MRL-lpr (The Jackson Laboratory). Mice of the first four strains were used at 8–10 weeks of age, while those of the latter strain were 12 weeks old. All animals were maintained under pathogen-free conditions in the animal facility of Università del Piemonte Orientale; they were fed ad libitum on rodent chow and water was freely available in the home cages, the ambient temperature was maintained at 21 ± 1 °C. All experimental procedures were conducted during the light phase of a 12:12-h light:dark cycle. Experimental procedures were conducted, following European guidelines, in accordance with both the University Ethical Committee and the National Institutes of Health Ministry and Care Committee (protocol number DB064.42), both of which approved the protocol.

Autoimmune Disease Mouse Models

Congenic control (C57BL/6 or B6), Mrl-lpr, Ctnnbfl/fl [25 (link)], and Lyz-M-Cre [26 (link)] mice were purchased from the Jackson Laboratory (Bar Harbor, ME) or Taconic Farms (Hudson, NY). B6.Sle1 [27 (link)], B6.Sle3 [28 (link)], B6.Yaa [29 (link)], and BWF1 [30 (link)] mice were bred in our mouse colony. Mice used for this study were 3 to 13 month females or—in the case of mice harboring the Yaa locus and their controls—males maintained in a specific pathogen-free environment. Mice were euthanized using a CO2 chamber and cervical dislocation. The Institutional Animal Care and Use Committee at the University of Texas Southwestern Medical Center approved all experiments on mice.

Mouse Strains for Lupus Research

Murine Models for Immune Regulation
