Pathway Agonist and Inhibitor Screening

The signalling pathway agonists/inhibitors Verteporfin (Tocris); iCRT3 (Sigma); DAPT (Merck Millipore) and GANT61(R and D Systems) were applied at the concentrations indicated during a 7-day incubation following 24-hour transduction of GSK3α/β floxed cells with Cre recombinase expressing lentivirus. Cells were stained with Hoechst (Sigma) at 1 µg/ml, imaged, using an IN Cell analyser 2200 (GE Healthcare) and cell number determined using IN Cell analyser workstation software (GE Healthcare).
Live cell imaging was carried out 5–7 days after viral transduction using an IncuCyte ZOOM cell imaging system (Essen Biosciences).

Culturing Breast Cancer Cell Lines

The SUM149, SUM159 and SUM190 cell lines were obtained from Asterand Inc (Detroit, MI) and cultured per manufacturer’s instructions and as described previously [35 (link)]. MDA-MB-231, SKBR3 cells and C3H10T1/2 cell lines were from American Type Culture Collection (ATCC) (Manassas, VA) and cultured per their instructions. Human mammary epithelial cells (HMEC) were obtained from Lonza Group Ltd. (Allendale, NJ) and cultured per manufacturer’s instructions using their Mammary Epithelial Growth Medium Kit (CC-3150). GANT58 (Cat # 3889), GANT61 (Cat # 3191), JK184 (Cat # 3341) and cyclopamine were obtained from Tocris/R&D Systems (Minneapolis, MN). GANT61 in gram quantities was from DC Chemicals (Shanghai, China). HPI-1/2/3/4 were a kind gift from Dr. James K. Chen, Stanford School of Medicine. HPI-1/3/4 were also purchased from Sigma Aldrich (St. Louis, MO). Cytochalasin D was from MP Biomedicals, LLC (Solon, OH). Hoechst-33342 and YOYO-1 dyes were from Thermo Fisher Scientific

Investigating Signaling Pathway Inhibitors in MCL

GANT61 Treatment of iKRAS Cells

Cells were treated with 5 μM of GANT61 (3191/10; R and D Systems) for 3–5 days prior to collection of RNA or protein lysates. Alternatively, iKRAS cells were plated at 150,000 cells with or without Dox (1 μg/mL) in the presence of absence of 5 μM GANT61 and RNA was collected 72 hr post-plating.
