Rare Earth Oxide Nanoparticle Synthesis

Rare earth oxides Y2O3 (99.99%), Yb2O3 (99.99%), and Er2O3 (99.99%) were obtained from China Rare Earth Online Co. Ltd. Rare earth chlorides (LnCl3, Ln: Y, Gd, Yb, Er) were prepared by dissolving the corresponding oxides in hydrochloric acid and then evaporating the water completely. Ammonium fluoride (NH4F, 98%), sodium hydroxide, hydrochloric acid, ethanol, methanol, methylene chloride, triethylamine (TEA), and dimethyl sulfoxide (DMSO) were purchased from Sinopharm Chemical Reagent Co., Ltd (China). Oleic acid (OA, 90%), 1-octadecene (ODE, 90%), and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) were purchased from Sigma-Aldrich. Anticancer drug doxorubicin hydrochloride (DOX, > 98%) was purchased from Shanghai Sangon Biotech Company (Shanghai China) and was deprotonated with addition of triethylamine to obtain the hydrophobic form. Carboxyl-terminated Poly(lactide-co-glycolide) (PLGA-COOH 50:50, Mw = 10000) was obtained from Jinan Daigang Bioengineer Company, China. Polyvinyl alcohol (PVA, Mw = 22000) was obtained from Kuraray CO. LTD, Japan. All reagents were used as received without further purification.

Synthesis and Characterization of Glucose-Functionalized Polymers

Monohydroxy-capped poly(ethylene glycol) (PEG113-OH) with a mean degree of polymerization, DP, of 113 (Mn = 5000 g/mol, Mw/Mn = 1.02) was purchased from Aldrich and dried at 30 °C under vacuum overnight prior to use. 2-Diethylaminoethyl methacrylate (DEA, Aldrich, 99%) and styrene (St, Aldrich, 99%) were passed through basic alumina column to remove inhibitor and then vacuum distilled from CaH2 and stored at − 20 °C prior to use. 2, 2-Bipyridine (bpy), copper bromide (CuBr), 2-bromoisobutyryl bromide, 2-aminoethyl methacrylate hydrochloride (AMA.HCl), d-glucono-δ-lactone, bortezomib (BTZ), and Alizarin Red S (ARS) were all purchased from J&K and directly used. Methanol, toluene, triethylamine (TEA), 1-methyl-2-pyrrolidinone (NMP), and isopropyl alcohol (IPA) were purchased from Sinopharm Group Chemical Reagent Co., Ltd., and then distilled from CaH2 under reduced pressure. Dichloromethane, chloroform, dimethyl sulfoxide (DMSO), and other organic solvents were purchased from Sinopharm Group Chemical Reagent Co., Ltd., and directly used without further purification. Bromide terminated poly(ethylene glycol) (PEG113-Br) macroinitiator and 2-gluconamidoethyl methacrylate (GAMA) monomer were prepared from PEG113-OH and 2-bromoisobutyryl bromide, and AMA.HCl and d-glucono-δ-lactone, respectively, according to literature procedures [35 (link), 36 (link)].

Synthesis of DOX-Loaded PEG-TMC Copolymer

All chemicals were obtained from the indicated sources. 1,3-Trimethylene carbonate (TMC) was purchased from Daigang Biomaterial Co. Ltd. (Jinan, China), and recrystallized twice with acetic ether and then dried 24 h in a vacuum before polymerization. Stannous octanoate (Sn(Oct)2) and methoxypolyethylene glycols (mPEG43) were purchased from Aldrich and used without purification. Doxorubicin hydrochloride (DOX·HCl) was purchased from Ark Pharm, Inc. (Arlington Heights, IL, USA) Triethylamine (TEA) was purchased from Sinopharm Chemical Reagent Co., Ltd. (Shenyang, China). Tetrahydrofuran (THF) and toluene were dried by treatment with Na and distilled before use. All other solvents and reagents used were purified by standard methods.

Synthesis and Purification of Functionalized Monomers

N-isopropyl acrylamide (NIPAM, J&K Scientific, 99.5%, Beijing, China) was recrystallized three times in n-hexane and methylbenzene to remove inhibitor. Diacetone acrylamide (DAA, Macklin Co., 99%, Shanghai, China) was recrystallized from n-hexane for three times to remove inhibitor. 2,2’-Azobis (isobutylronitrile) (AIBN, J&K Scientific, 98%, Beijing, China) was recrystallized from ethanol. N-acryloyl-3-aminophenylboronic acid (AAPBA) and chain transfer agent S-1-Dodecyl-S′-(R,R′-dimethyl-R′′-acetic acid) trithiocarbonate (DDMAT) was synthesized and purified according to the literature procedures [36 (link),37 (link)]. Acryloyl chloride, aminophenylboronic acid (APBA), adipic dihydrazide (ADH), poly(vinyl alcohol) (PVA, polymerization degree: 1750, hydrolysis degree: 87-89%) were purchased from J&K Scientific, Beijing, China. Sodium phosphate dibasic, sodium dihydrogen phosphate, methanol, dimethyl sulfoxide (DMSO), triethyl amine (TEA) and acetone were purchased from Sinopharm Chemical Reagent Co. Ltd., Beijing, China.

Male ICR mice (SPF, 18–22 g) were obtained from Animal Center of Anhui Medical University (Hefei, China).
