Neuroblastoma and Microglia Cell Experiments

Human neuroblastoma SH-SY5Y cells and mouse microglia BV2 cells were provided by Institute of Basic Medical Sciences (IBMS), Chinese Academy of Medical Sciences and Peking Union Medical College (CAMS and PUMC) (Beijing, China). Dulbecco’s modified eagle’s medium (DMEM) and fetal bovine serum (FBS) were bought from Thermo Fisher Scientific, Inc. (Waltham, MA, United States). 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazoliumbromide (MTT), KCl, hydrogen peroxide (H2O2), L-glutamic sodium (MSG), sodium nitroprusside dihydrate (SNP), okadaic acid (OA), amyloid β-protein fragment 1–42 (Aβ), and lipopolysaccharides (LPS) were purchased from Sigma-Aldrich (St. Louis, MO, United States). L-Arctigenin, Dihydrocapsaicin, Rhapontigenin, 6-Shogaol, 6-Gingerol, 10-Gingerol, Isorhapontigenin, Nonivamide and Flavokawain B were bought from Target Molecule Corp. Demethoxycurcumin, Bisdemethoxycurcumin, Carnosol, Cardamonin, Matairesinol, Bavachromene, 3,4-Dimethoxycinnamic acid were purchased from Yuanye Bio-Technology (Shanghai, China). NO assay kit was acquired from Beyotime Institute of Biotechnology (Shanghai, China).

Characterization of Cardamonin Compound

Reagents of analytical grade were used as received from their commercial sources, while cardamonin (98% purity) was obtained from Shanghai Yuanye Bio-Technology Co. Ltd (Shanghai, China). Elemental analysis was performed on a Vario MACRO CUBE CHNS analyzer, while thermogravimetric analysis (TGA) was performed via a Mettler Toledo TGA analyser up to 800 o C with a heating rate of 10 o C/min in an atmosphere of N2. IR spectra were recorded by a Perkin Elmer FTIR spectrophotometer within the range (4000-400 cm -1 ) using KBr discs while melting points have been measured using an Electrothermal IA9100 digital m.p. apparatus measuring within a range of (0 o C-400 o C). HPLC purity analysis was performed using an Agilent 1260 Infinity system equipped with a Zorbax 300SB-C18 column (5 µm, 250 mm x 4.6 mm). Mobile solvent A was water, while B was acetonitrile and the flow rate used was 1.25 ml/min The time program used for the HPLC analysis was 55% B (0-5 min), 55% -85% B (5-25 min) and 99% B (25-30 min).
1 H NMR spectra were obtained via a Bruker FT-NMR 300 MHz, 400 MHz and 600 MHz. TLC was performed using 60F aluminium silica gel aluminium plates with 254 nm fluorescent indicator from Merck. UV-Vis spectra were obtained via an UltroSpec 8000 spectrophotometer.
