Extracellular Vesicle miRNA and mRNA Profiling

Total RNA was isolated from EV pellets collected from donor plasma using miRNeasy kit (Qiagen). Seventy-five nanograms of RNA were reverse transcribed using miScript RT II kit (Qiagen) using HiFlex buffer to amplify miRNA and mRNA concurrently. Expression of miR-24, miR-30b-5p, and miR-142–3p (primers purchased from Qiagen), PD-L1 and β-actin (primers purchased from Sigma) was assessed by real-time quantitative PCR, and average Ct values were plotted.

miRNA Transfection in Immune Cells

MiR-24, miR-30b, and miR-142-3p mimics or inhibitors were purchased from Qiagen (Gaithersburg, MD, USA). As a negative control, miRNA mimics with no known human targets was used (Qiagen). Transient transfection of miRNA mimics, inhibitors or control was performed using Lipofectamine 2000 (Invitrogen, Carlsbad, CA). MΦ were transfected at a final concentration of 50 nM oligos while DC were transfected with 100 nM oligos. As a positive transfection control, we used DY-547 labelled siRNA (Life Technologies).

Overexpressing miRNA in Differentiated Macrophages and Dendritic Cells

Transient miRNA overexpression was performed by transfecting miRNA mimics using Lipofectamine 2000 (Invitrogen-Life Technologies Corporation, Carlsbad, CA, USA) according to the manufacturer’s instructions. Day 7-differentiated MΦ or DC were transfected at a final concentration of 50 nM. Red siGLO oligos (ThermoFisher Scientific, Grand Island, NY, USA) were used to determine transfection efficiency.
