Purification and Characterization of Crocin and Safranal

Commercial samples of crocin (crocin-1, crocetin di-gentiobiose ester) and safranal (488%) were purchased from Sigma-Aldrich (Milan, Italy). safranal was further purified by column chromatography on silica gel (230-400 mesh, G60 Merck, ethyl acetate:n-hexane 1:3). The other compounds (3-9) have been synthesized and characterized to ensure purity as reported in the literature 29 .

Saffron Compound Preparation Protocol

The HCT116 cells were cultured in the RPMI 1640 Medium (Hyclone, Marlborough, MA, USA) while HCT116 +3, HCT116 +5, and HCT116 +3+5 cells were cultured in McCoy’s Medium (Hyclone, Marlborough, MA, USA). Both media were supplemented with 10% fetal bovine serum (Sigma Aldrich, St. Louis, MO, USA) and containing 1% of 100 U/mL penicillin and 100 µg/mL streptomycin (Sigma Aldrich, St. Louis, MO, USA) at 37 °C in a humidified 5% CO2 atmosphere. Cells were sub-cultured every 3–5 days using Trypsin 0.25% (Hyclone, Marlborough, MA, USA).
Saffron, safranal, and crocin preparations: Saffron crude extracts (Gulf Pearls SPRL Brussels, Belgium, www.gp-food.com, accessed on 3 May 2021) and specific compounds (safranal and crocin (Sigma Aldrich, St. Louis, MO, USA)) were used as previously described [7 (link),17 (link)]. Saffron was grinded by a pestle and mortar and dissolved in water at a final concentration of 20 mg/mL and mixed on an orbital shaker in the dark for 1 h then used at different concentrations.

Saffron Extract Standardized for Clinical Trial

The saffron (Crocus sativus L.) stigmas were extracted in the factory of Pharmactive Biotech Products S. L. in Madrid (Spain) to produce the commercial saffron extract, branded as affron®, and dosed in tablets as preferred galenic form for the in vitro digestion and the clinical trial. Each tablet contained 14 mg of dried affron® standardised to ≥3.5% lepticrosalides®, defined as the content of bioactive compounds including Safranal and crocin by HPLC [12 (link), 13 (link)], together with the following excipients: microcrystalline cellulose, calcium hydrogen phosphate, povidone, croscarmellose sodium, colloidal anhydrous silica, and magnesium stearate as excipients and coated with hypromellose (HPMC), macrogol (PEG) 8000, and carnauba wax.
Safranal, crocetin, p-nitroaniline, and digestive enzymes were purchased from Merck (Darmstadt, Germany). Kaempferol was purchased from Extrasynthese (Genay, France), and trans-crocin-4 was purchased from Phytolab (Vestenbergsgreuth, Germany). All the solvents used for the chromatographic analyses were of HPLC grade and purchased from Merck and VWR (Pennsylvania, United States).

Saffron Characterization and Aroma Evaluation

Samples. Saffron from the Protected Designation of Origin (PDO) “Azafrán de La Mancha” was directly purchased from the producer Agrícola Técnica de Manipulación y Comercialización (Minaya, Albacete, Spain). Category I saffron was ground and characterized according to ISO 3632:2011 [25 ] ( A1cm1% 440 nm: 235 ± 2, A1cm1% 257 nm: 97 ± 3, and A1cm1% 330 nm: 22 ± 1). Cured ham was obtained from the company Benibaldo, S.A.U. (Albacete, Spain), and rice flour obtained from Panificadora Conquense Agrícola, S.A. (Cuenca, Spain), a usual product in the ham industry, was used in this study as a solid support to retain the patterns and for solid dilution of saffron.
Patterns. A total of four patterns of the major aromatic components of saffron were used. Safranal (purity: 90%), linalool (purity: 97%), isophorone (purity: 97%), and 2,6,6-trimethyl-2-cyclohexene-1,4-dione (purity: 98%) were obtained from Sigma-Aldrich (Madrid, Spain).
Solvent. Cyclohexane of analytical grade was obtained from Panreac (Barcelona, Spain).

Saffron Extracts and Compounds Effects

The stigmas of C. sativus L. was purchased from Novin Saffron Co. (Mashhad, Iran) and analyzed in accordance to ISO/TS 67321–2. Aqueous and ethanolic extracts were extracted from saffron in our laboratory and administered at the doses of 40, 80, and 160 mg/kg, IP. Safranal (0.1, 0.3 and 0.5, ml/kg, IP) was obtained from Sigma-Aldrich (St. Louis, MO, USA). Dosage selection was based on our previous study (12 (link)). Harmaline was purchased from Fluka Chemie GmbH; Buchs, Switzerland and administered at the dose of 30 mg/kg via intraperitoneal rout (25 (link)). Propranolol was donated by Tehran Darou Pharmaceutical Co. (Tehran, Iran) and administered at the doses of 1, 2, and 5 mg/kg, IP (26 (link)). Both extracts and propranolol were prepared freshly in normal saline (0.9%). Paraffin was used as a vehicle for Safranal. All compounds were administered at the intraperitoneal rout.

and ethylenediaminetetraacetic acid (EDTA) were procured from Loba
Chemie and SD Fine Chemicals, respectively. All other chemicals/reagents
used were of research grade. Ultrapure water from Direct-Q3 water
purification system (Merck-Millipore) was used for all experimentations.
