Synthesis of Fluorescent Polymer Hydrogels

N-vinylcaprolactam (VCL) and acetoacetoxyethyl methacrylate (AAEM) were obtained from Sigma Aldrich (St. Louis, MO, USA), vacuum distilled under nitrogen and purified by conventional methods. The fluorescent marker rhodamine B, the initiator 2,2-azobis-2-methylpropionamidine dihydrochloride (AMPA) and the cross-linker N,N-methylene bisacrylamide (MBA) were purchased from Sigma Aldrich and used as received. Deionized water was employed as synthesis medium and solvent. Toluene for cleaning cover slides and ethanol were bought from Sigma Aldrich (anhydrous, 99.8%). A Sylgard 184 elastomer kit, made up of Sylgard 184 monomer and Sylgard 184 base, was obtained from Dow Corning (Midland, MI, USA).

Synthesis and Purification of OFP Crosslinker

2,2’- Azobis(2-methylpropionitrile) (AIBN) (Sigma-Aldrich) was purified by recrystallization from methanol prior to use. N-Vinylcaprolactam (VCL) (Sigma-Aldrich) and tetrahydrofuran (Fisher) was distilled before reaction. Potassium ethyl xanthate, 2-bromopropionyl bromide, diethyl ether, hexane, dichloromethane, calcium sulfate anhydrous, and dialysis tubing (MWCO 5 kDa) (Fisher) were used as received. Bis(hydroxyalkyl) poly(dimethylsiloxane) (PDMS65, Mn = 5600 Da) was purchased from Sigma-Aldrich. Triflic acid was purchased from Sigma-Aldrich.

Synthesis and Characterization of PDMS-g-PVCA Copolymers

2,2′-Azobis(2-methylpropionitrile) (AIBN) (Sigma-Aldrich, Saint Louis, MO, USA) was recrystallized from methanol before use. N-Vinylcaprolactam (VCL) (Sigma-Aldrich) and tetrahydrofuran (THF; Fisher) were distilled prior to use. Potassium ethyl xanthate, 2-bromopropionyl bromide, diethyl ether, hexane, dichloromethane, calcium sulfate anhydrous, sodium bicarbonate, monobasic and dibasic sodium phosphate, and dialysis tubing (MWCO 3 kDa and 20 kDa) were from Fisher Scientific (Suwanee, GA, USA) and were used as received. Poly(dimetylsiloxane) bis(hydroxyethyloxypropyl) (PDMS65, Mn = 5600 Da), 2-methyl-2-oxazoline (MOXA), trifluoromethanesulfonic anhydride (Tf2O), potassium hydroxide, calcium chloride, diethyl ether, and methanol were purchased from Sigma-Aldrich. Potassium carbonate, sodium bicarbonate, sodium chloride, chloroform, hydrochloric acid, and glass fiber filters G4 were purchased from Fisher Scientific. Diethoxydimethylsilane, 1,3-bis(4-hydroxybutyl)tetramethyldisilane were from Silar Laboratories (Riegelwood, NC, USA). Doxorubicin hydrochloride (DOX) was purchased from LC Laboratories. Deionized (DI) water with an 18.2 MΩ cm resistivity was used for the preparation of aqueous solutions.

Polymeric Drug Delivery System Synthesis

Emulsifier: sodium dodecyl sulfate (SDS; STEOL®CS-230 Stepan). Buffer: sodium hydrogen carbonate (Sigma-Aldrich, ≥99.7%) used as provided. Synthesis and Characterization of VCL-Based Hydrogels

N-Vinylcaprolactam (VCL, 98%; Sigma-Aldrich) was purified by distillation under vacuum. 2-Methoxyethyl acrylate (MEA, 98%; Sigma-Aldrich) was passed through a basic aluminum oxide column before use to remove the inhibitor. The initiator 2,2 0 -azobis(2-methylpropyonamidine)dihydrochloride (AMPA, 97%; Sigma-Aldrich), the crosslinker N,N 0 -methylenebisacrylamide (BIS, 99%; Sigma-Aldrich), chloroform (99%; VWR), deuterated water (D 2 O, 99.9%; Deutero) and the model drug Cyanine3 (Cy3, Lumiprobe) were used as received.
