Somatostatin

Manufactured by GeneTex
Sourced in United States

Somatostatin is a hormone produced in the hypothalamus and pancreas that regulates the secretion of other hormones, such as growth hormone and insulin. It plays a role in inhibiting the release of these hormones, thereby helping to maintain homeostasis in the body.

Immunofluorescent Pancreatic Cell Staining

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mouse pancreata were fixed in 4% paraformaldehyde and embedded with paraffin. Rehydrated 5 μm sections were stained with primary antibodies against insulin (dilution 1:1000; Dako, Santa Clara, CA, USA), somatostatin (dilution 1:300, GeneTex., Irvine, CA, USA), and glucagon (dilution 1:100; Abcam, Cambridge, UK) followed by secondary antibodies (dilution 1:500; anti-guinea pig Alexa Fluor 488, dilution 1:500; anti-mouse, Alexa Fluor 647, and dilution 1:500; anti-rabbit Alexa Fluor 546) as previously described (Vierra et al., 2015 (link)). Sections were imaged either with a Nikon Eclipse TE2000-U microscope or fluorescent ScanScope (Aperio).

Mouse pancreata were fixed in 4% paraformaldehyde and embedded with paraffin. Islet and β-cell Imaging with Confocal Microscopy

Check if the same lab product or an alternative is used in the 5 most similar protocols
Islet and β-cell imaging was performed with a Leica laser scanning confocal microscope system (DMIRE2; Leica Microsystems, Wetzlar, Germany) with ×20 (numerical aperture 0.75) and ×60 (numerical aperture 1.4) oil immersion objectives as previously described (17 (link)). Primary antibodies include insulin (guinea pig, 1:200; Dako Diagnostics, Mississauga, Ontario, Canada), glucagon (mouse, 1:200; NovoCastra Laboratories, Leica Microsystems), and somatostatin (rabbit, 1:200; GeneTex Inc., Irvine, CA) and rabbit anti–Syt-7 antibody (1:200; Synaptic System). Deconvolution algorithm was applied to images to remove out-of-focus background noise. Colocalization was quantified using Volocity software (PerkinElmer, Waltham, MA). Regions of interest (ROIs) were outlined; the number of colocalized pixels within a particular ROI was determined and expressed as percentage of total pixels within that ROI.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
