Curcumin Cytotoxicity on Prostate Cells

PCa cell lines (PC3, DU145, LNCaP, and 22RV1) and normal prostate cells (RWPE-1) were purchased from Shanghai Zhongqiao Xinzhou Biotechnology Co., Ltd. The culture conditions of PC3, DU145, LNCaP, and RWPE-1 cells were consistent with previous reports [15 (link)]. 22RV1 cells were cultured in RPMI-1640 supplemented with 10% fetal bovine serum (FBS) and 1% penicillin/streptomycin. Cur (0, 25, 50, and 100 μg/ml) was used to treat RWPE-1 and PCa cells for 48 h.

Establishing Bicalutamide-Resistant Prostate Cancer Cells

Human prostate cancer cells (PC3, DU145, LNCap) are purchased by two agent companies from the Cell Resource Center of Shanghai Academy of Life Sciences, Chinese Academy of Sciences. The PC3 and LNCap cells were purchased from Shanghai Zhong Qiao Xin Zhou Biotechnology Company. The DU145 cells were purchased from Shanghai Biowing Applied Biotechnology Company. bicalutamide-resistant (Bica-R) cells was constructed early in our group [33 (link)]. The LNCap cells, one of the androgen-dependent prostate cancer cell strains, were treated with bicalutamide (10 μM) (Selleck Chemicals, Houston, TX, USA), respectively for at least 6 months to generate bicalutamide-resistant cells. Cells were cultured in RPMI-1640 (Gibco, USA) medium supplemented with 10% fetal bovine serum (Gibco, USA), 1% penicilin/streptomycin ((Life Technologies, Carlsbad CA) in a humidified incubator of 5% CO2 at 37 °C.

Culturing Prostate Cancer Cell Lines

The RWPE-1, LNCaP, 22RV1, PC3, and DU145 cell lines were obtained from Zhong Qiao Xin Zhou Biotechnology Co., Ltd. (Shanghai, China). HEK-293T cells were purchased from Kunming Cell Bank (Kunming, China). The cells were cultured and aliquots were stored in liquid nitrogen for future use. RWPE-1 cells were cultured in customized medium (ZQXZ Bio, Shanghai, China). LNCaP, 22RV1, and DU145 cells were cultured in RPMI-1640 medium supplemented with 10% fetal bovine serum (FBS) and 1% penicillin–streptomycin (P/S) at 37 °C in a 5% CO2 environment. PC3 cells were cultured in ATCC-formulated F-12K medium supplemented with 10% FBS and 1% P/S at 37 °C in a 5% CO2 environment. HEK-293T cells were cultured in DMEM/HIGH GLUCOSE (HyClone, Logan, Utah, USA) supplemented with 10% FBS and 1% P/S at 37 °C in a 5% CO2 environment. Olaparib (AZD2281) and ML216 (CID-49852229) were purchased from MedChemExpress (Monmouth Junction, NJ, USA).

Bicalutamide Cytotoxicity in Prostate Cells

The RWPE-1 and PCa cell lines (LNCaP, PC-3, and DU145) were purchased from Shanghai Zhong Qiao Xin Zhou Biotechnology (Shanghai, China). LNCaP and PC-3 cells were cultured in RPMI-1640 medium (Gibco, USA) supplemented with 10% fetal bovine serum (FBS; Procell, Wuhan, China). RWPE-1 and DU145 cells were cultured in a special medium purchased from Shanghai Zhong Qiao Xin Zhou Biotechnology. All cell lines were incubated at 37℃ in a humidified cell incubator set to 5% CO2. Bicalutamide concentrations ranged from 2.5 to 12.5µM.

Prostate Cell Culture and Drug Treatment

LNCaP cells were treated with ENZ, BIC, and ARN solution (MCE, Shanghai, China) for 24 h to simulate drug therapy, and then collected for further experiments. RWPE-1 cells were cultured in special medium (ZQ-1303) and purchased from Shanghai Zhong Qiao Xin Zhou Biotechnology Co., Ltd. (China).

Prostate Cancer Cell Lines Transfection

PCa (PC3, DU145, LNCaP, 22RV1) and normal control cells (WPMY-1) were purchased from Zhong Qiao Xin Zhou Biotechnology (Shanghai, China). PC3 and DU145 cells were cultured in F-12K medium, LNCaP and 22RV1 cells in 1640 medium, and WPMY1 cells in DMEM medium. Complete medium was obtained by adding 10% fetal bovine serum (FBS, ScienCell, USA) and 1% penicillin and streptomycin. The cell incubator was set at 37°C and 5% CO. The small interfering RNAs (si-RNAs) for cell transfection and the transfection kit were purchased from RiboBio company (Guangzhou, China). The transfection procedure was conducted according to the manufacturer’s instructions.

Prostate Cancer Cell Line Cultivation

A human prostatic stromal myofibroblast cell line (WPMY-1) and prostate cancer cell lines (PC3, 22RV1, and LNCap) were purchased from the Zhong Qiao Xin Zhou Biotechnology Co., Ltd. All culture media were supplemented with 10% FBS (04-001-1ACS; BI, Beit HaEmek, Israel), 1% penicillin (100 U/mL), and streptomycin (100 g/mL) solution (SV30010; HyClone, Logan, UT, USA).
BALB/c Nude mice (4 to 6 weeks old) were purchased from Hua Fu Kang Biotechnology Company (Beijing, China) and were reared according to the Laboratory Animal Welfare Ethics Committee of Zunyi Medical University.

Prostate Cancer Tissue Collection

Sixteen pairs of carcinoma and adjacent tissues were collected from Zunyi Medical University according to ARRIVE guidelines. Ethical approval for this work was granted by the Institutional Review Board of Zunyi Medical University.
