Protopanaxadiol Formulation Development

20(S)-Protopanaxadiol (PPD) (purity>98.0%) was obtained from Xian Plant Bio-Engineering Co., Ltd. (Shaan Xi, China). PEG-8 Caprylic/Capric Glycerides (LAS), Labrafac CC and Lauoroglycol CC were obtained from Gattefosse Co. (Saint Priest, Cedex, France). Capmul MCM EP was obtained from ARITEC Co. (Columbus, OH, USA). Miglyol 812 N and Poloxamer 188 were gifts from OLEO Chemicals (Witten, Germany) and BASF Co. (Ludwigshafen, Germany), respectively. Limonene, Tween 20, Tween 80, polyethylene glycol 400 (PEG400) and isopropyl myristate were obtained from Sigma-Aldrich Co. (St. Louis, MO, USA). Cetyl palmitate was obtained from TCI (Tokyo Kasei, Japan). Strat-M™ was purchased from Merck Millipore (Billerica, MA, U.S.A.). HPLC-grade water, acetonitrile, and methanol were obtained from Thermo Fisher Scientific Co. (Pittsburgh, PA, USA).

Formulation and Characterization of Topical Antimicrobial Agents

DMEM (Dulbecco’s Modified Eagle’s medium) was purchased from ThermoFisher, MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) from Sigma-Aldrich (St. Louis, MO, USA) and DMSO (99% dimethyl sulfoxide) from Sigma-Aldrich (Barcelona, Spain).
Mueller–Hinton broth (MHB), Brain Heart Infusion (BHI), Clostridium reinforced medium (CRM), Tryptone Soy Agar (TSA), and Sabouraud Dextrose Agar (SDA) were purchased from Oxoid (Basingstoke, UK). Beren (cosmetic diluent) was acquired from Scharlab (UK).

Pranoprofen Topical Formulation Development

Pranoprofen (2-(5H-chromeno [2,3-bpyridin-7-yl) propanoic acid) was a kind gift from Alcon Cusi (Barcelona, Spain). Tween® 80 (Polyethylene glycol sorbitan monooleate) and Castor oil (Ricinus communis L.) were acquired from Sigma-Aldrich Química (Barcelona, Spain). Lanette® 18 (stearyl alcohol) was purchased from Cognis (Dusseldorf, Germany). PEG-8 Caprylic/Capric Glycerides (LAS) was received as a gift sample from Gattefosse (Saint-Priest, France). The tattoo ink (Viking-Ink BW Black Tribal, Náquera, Spain) was purchased from Viking-Ink B&W. The MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) utilized for cell viability was obtained from Invitrogen Alfagene® (Carcavelos, Portugal), and the HaCat was obtained by Cell Lines Service (CLS, Eppelheim, Germany). MilliQ water obtained by a MilliQ® Plus System lab and was used across formulations and all experiments. The phosphate buffer saline (PBS) tablets pH 7.4 were acquired from Sigma (Darmstadt, Germany) and prepared as according to the manufacturer’s instructions. All the other chemicals and reagents used in this investigation were of analytical grade.

High-performance liquid chromatography (HPLC)- grade methanol, acetonitrile, and water were purchased from Thermo Fisher Scientific Co. (Pittsburgh, PA, USA). Female human cadaver skin (2 × 2 cm2) was purchased from a skin and tissue bank (Hans Biomed Co., Seoul, Korea). Studies that use human cadaver skin from a tissue bank do not require approval from the Institutional Review Board (IRB) or ethics committee (Article 33, Enforcement Rule of the Bioethics and Safety Act in the Republic of Korea) [22 (link)].

The purified water utilized in all experiments was collected from a MilliQ® Plus System (Darmstadt, Germany), lab-supplied. The tablets of phosphate-buffered saline (PBS) were acquired from Sigma-Aldrich Chemie (Steinheim, Germany) as well as processed as specified by the manufacturer and stored in fridge for further use. All other chemicals for analytical experiments and reagents utilized in the research were of analytical or high-performance liquid chromatography (HPLC) grade and acquired from Fisher Scientific (Leicestershire, UK).
