Sodium hydroxide, potassium phosphate monobasic and potassium phosphate dibasic and EDTA were purchased from J.T. Baker (Center Valley, PA, USA). HPLC-grade acetonitrile and methanol were purchased from Thermo Fisher Scientific (Waltham, MA, USA). Ultra-pure water was obtained using a Milli-Q® Q-POD Millipore System (EMD Millipore, Burlington, MA, USA).

Quantitative Analysis of Kratom Alkaloids

Acetonitrile and methanol were purchased from Fisher Scientific (Fair Lawn, NJ, USA). Blank human plasma (K2EDTA, mixed biological sex, pooled, lot HMN99236) and whole blood (K2EDTA, mixed biological sex, pooled, lot HMN541526) were purchased from BioIVT (Westbury, NY, USA). All other chemicals and reagents were analytical grade.

Methanol (MeOH) was purchased from Pharmco-Aaper (Shelbyville, KY) and Fischer Scientific (Waltham, MA). UPLC-grade water (H2O) and acetonitrile (CH3CN) were purchased from Fisher Scientific. Pooled human intestinal microsomes (HIM) (n = 18 donors) were purchased from Xenotech (Lenexa, KS).

Quantification of Pharmaceutical Compounds in Serum

LC-MS grade acetonitrile and methanol were purchased from VWR (Radnor, PA), and LC-MS grade formic acid was purchased from Thermo Scientific (Rockford, IL). Water was prepared using an Elga Purelab Ultra (Siemens Water Technologies, Woodridge, IL) water purification system.
Pooled human serum and serum from individual donors were purchased from Valley Biomedical Products & Services (Winchester, VA), Innovative Research (Novi, MI), Sigma-Aldrich (St. Louis, MO), and BioIVT (Westbury, NY). Serially drawn serum samples from women who had been administered an oral cocktail of 100 mg Caffeine, 250 mg tolbutamide, 30 mg dextromethorphan, and 2 mg alprazolam were obtained from the UIC/NIH Center for Botanical Dietary Supplements Research (registered with ClinicalTrials.gov as , UIC Institutional Review Board #2015-0651). These serum samples contained no patient identifying information. All serum was stored at −80 °C until use.

Intramuscular Alprazolam Administration

Alprazolam was purchased commercially (Sigma-Aldrich, MO, US) and was dissolved in 100% propylene glycol and diluted in water. Final solutions were prepared using 50–80% propylene glycol and 20–50% sterile water. Solutions were prepared immediately before injections, and all injections were administered intramuscularly. Propylene glycol may cause lesions if given intramuscularly at high concentrations/volumes. Therefore, for each dose, we used the highest concentration of drug and smallest amount of propylene glycol/injection volume possible that still maintained the drug in solution (injection volume varied between 0.01–0.04 ml/kg). Vehicle injections were given at the highest concentration of propylene glycol (80% propylene glycol and 20% sterile water), at the highest injection volume administered for that concentration (0.04 ml/kg).
