Plan apo λ 100 oil objective

Manufactured by Nikon
Sourced in Japan

The Plan-Apo λ 100× oil objective is a high-magnification objective lens designed for use in microscopy applications. It provides a magnification of 100× and is optimized for use with oil immersion. The lens is engineered to deliver a flat, distortion-free image across the entire field of view.

Superresolution Imaging of Whole Cells

SIM imaging was performed on a W1 spinning-disk microscope (CSU-W1; Nikon, Japan) combined with the Live-SR system (Roper Scientific) and equipped with a Plan-Apo λ 100× oil objective (1.45 NA; Nikon, Japan). Excitation and band-pass filters were as follows: DAPI was imaged with a 405-nm laser and emission of 445/45 nm, Alexa Fluor 488 was imaged using 488-nm excitation and emission of 525/30 nm, and Alexa Fluor 568 was imaged using 561-nm excitation and emission of 617/73 nm. Three-dimensional (3D) image stacks of whole cells were acquired with a scalable complementary metal-oxide semiconductor (sCMOS) camera (Prime 95B; Photometrics, USA), using an image pixel size of 78.96 nm by 78.96 nm (1,200 by 1,200 pixels) and an 0.2-μm z-stack step size. The raw images were processed to superresolution images by using the Live-SR algorithm reported by York et al. (21 (link)).

Spinning Disk Confocal Microscopy Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
All cells were imaged on a spinning disk confocal microscope except for Figure 3, which was on a Zeiss LSM 880 Confocal Laser Scanning Microscope equipped with a Plan-Apochromat 63× oil objective (1.4 N.A) and Airyscan detector and controlled by Zen Black (Zeiss). The spinning disk is a Nikon Ti2E body with PSF; CSU-X1 Yokogawa confocal scanner unit; OBIS 405, 488, 561, and 640 lasers and an ANDOR iXON Ultra 512X512 EMCCD camera. Images were acquired with Nikon Plan Apo λ 100× oil objective (1.4 N.A). Images were acquired with micro-manager 2.0. Images were processed in Fiji (NIH) and Adobe Illustrator (Adobe).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
