Other solvents were purchased from Innochem (Beijing, China) and meet or exceed the analytical grade standard. The structures of all pure lipids are shown in Fig. S1 and S2. Anthraquinone was purchased from Rhawn (Shanghai, China).

Blood Metabolites in Aging Hens

Overnight fasting blood samples were collected from 6 randomly selected hens in each treatment group, consisting of hens at 58 and 65 weeks of age from a wing vein. On those same days, a second blood sample was collected 30 min after re-feeding 20 g of feed. Commercial kits were used to determine plasma triacylglycerol (TG, Sigma-Aldrich, St. Louis, MO, USA), glucose and NEFA (non-esterified fatty acid, Wako Chemicals, Osaka, Japan) levels. Plasma insulin and interleukin-1β (IL-1β) concentrations were determined by a validated commercial ELISA kit (Cat.# 10-1249-01, Uppsala, Sweden, KMC006; Biosource International, Camarillo, CA, USA, respectively) [28 (link),29 (link)].

Lipid Profiling Internal Standards

For the authentic internal standards (IS), triacylglycerol (TG) 11:0/11:0/11:0 and free fatty acid (FFA) 17:0 were obtained from Sigma-Aldrich (St. Louis, MO, USA), and phosphatidylcholine (PC) 13:0/13:0, phosphatidylethanolamine (PE) 15:0/15:0, phosphatidylinositol (PI) 8:0/8:0, lysophosphatidylcholine (LPC) 15:0, lysophosphatidylethanolamine (LPE) 13:0, and lysophosphatidylinositol (LPI) 13:0 were purchased from Avanti Polar Lipids (Alabaster, AL, USA). Cardiolipin (CL) 15:0/15:0/15:0/15:0 was synthesized in our laboratory previously [16 (link)]. Other chemicals and reagents, which were of the highest grade available, were purchased from Sigma-Aldrich unless specified. The mixed solution of all the IS was prepared with methanol (containing 0.006% butylated hydroxytoluene, w/v) and stored at −80 °C for no more than two weeks before use.

Lipidomics Standards Initiative Protocol

HPLC-grade methanol, acetonitrile (ACN), and isopropanol (IPA) were purchased from Merck (Darmstadt, Germany), HPLC-grade tert-butyl methyl ether (MTBE) and ammonium acetate from Sigma−Aldrich (St. Louis), HPLC-grade chloroform (CHCl3) from Duksan (Ansan-si, South Korea), and ultrapure water was prepared by a Milli-Q system (Millipore). All internal standards were purchased from Avanti Polar Lipids (Alabaster), except FA 22:0-d4 (ten Brink, Amsterdam, The Netherlands) and triacylglycerol (TG) 15:0/15:0/15:0 (Sigma-Aldrich).
