LCMV infection in C57BL/6 mice

C57BL/6 mice (H-2b) were originally obtained from Charles River Laboratories and further bred in the animal facilities of the University of Konstanz. Mice were kept under pathogen-free conditions with a light-dark cycle of 12 h and used at 8–12 weeks of age, at a weight of 20–25 g, in accordance with the rules of the authority of Regierungspräsidium Freiburg (G-18/10). The number of mice for each set of experiments was determined using G*Power 3.0.10 (comparison of two independent groups, effect size d: 0.79, α err prob: 0.05 [25 (link)]. Food and water were provided ad libitum. Mice were infected with 200 pfu LCMV-WE i.v. CB-5083 was dissolved in 0.5% methyl cellulose in water and administered by oral gavage (50 mg/kg) [19 (link), 20 (link)]. Control groups (vehicle) were treated with 0.5% methyl cellulose in water. Mice were randomly allocated to experimental groups and no blinding method was used. There was no animal exclusion criteria. Animal experiments were conducted in full accordance with the ARRIVE guidelines.

Isolation of Murine Peritoneal Exudate Cells

C57BL/6 mice (H2b, aged 8–20 weeks) were obtained from Charles River (Sulzfeld, Germany) and housed under SPF conditions in individually ventilated cages (IVCs) at the animal facility of the DKFZ. C57BL/6 mice were injected intraperitoneally (i.p.) with 1 ml of 3% (w/v) thioglycolate containing medium (Applichem, Darmstadt, Germany; Cat. No. A3869) using a 27 G needle. Four days later, mice were sacrificed by gradual CO2 exposition and PECs were isolated upon lavage of the peritoneum with 8 ml ice cold PBS. This procedure yielded approximately 1.5 x 107 PECs per mouse.

Generation and Characterization of ApcMin/+ LMP7-/- Mice

C57BL/6 mice (H-2b) were obtained from Charles River (Sulzfeld, Germany). LMP7-/- mice were kindly provided by Dr. John J. Monaco (Department of Molecular Genetics, Cincinnati Medical Center, OH). C57Bl/6J-ApcMin/+[CD44] mice on a pure C57BL/6 background were obtained from Prof. Jan Paul Medema (University of Amsterdam, The Nederlands). LMP7-/- mice were bred with ApcMin/+ mice to obtain ApcMin/+ LMP7-/- mice. Animals were housed in an air-conditioned, accredited animal facility on a 12 hour light/dark cycle and fed standard, autoclaved laboratory animal diet and water ad libitum. Genotypes of mice were verified via PCR using genomic DNA extracted from tail biopsies to determine knockout and Apc heterozygous state. Investigations have been conducted in accordance with ethical standards, the Declaration of Helsinki, German and EU guidelines and have been approved by the animal experimentation review board of Regierungspräsidium Freiburg.

Murine Macrophage and Fibroblast Isolation

Media and supplements were purchased from Invitrogen-Life Technologies. Recombinant murine IFN-␥ and TNF-␣ was purchased from Peprotech and used at 200 U/ml and 100 U/ml, respectively.

C57BL/6 Mouse Immunization Protocol
