Western Blot Analysis of Nuclear Proteins

Sucrose cushion-purified Nuclear proteins were used 49 (link). For Western blots, equal amounts of protein were resolved by 4–20% SDS-PAGE (Genscript) and transferred to PVDF membranes (Millipore). The membranes were blocked for 1 h at room temperature with Odyssey blocking buffer (LI-COR) and incubated overnight at 4 °C with primary antibodies. After incubation with IRDye 680/800-conjugated secondary antibodies, the membranes were scanned using the Odyssey Infrared Imaging system (LI-COR). The following antibodies were used: IRF1 (sc-497, Santa Cruz), IRF7 (sc-13041, Santa Cruz), ZEB1 (sc-25388, Santa Cruz), ZEB2 (sc-271984, Santa Cruz), α-Tubulin (A01622, Genscript), and LaminB2 (MAB3536, Chemicon).
