Rotavirus Infection in Genetically Modified Mice

B6129SF2/J wild-type and Mavs-/- mice (B6;129-Mavstm1Zjc/J) were purchased from the Jackson laboratory (stock number: 101045 and 008634 respectively) and bred in-house at the Veterinary Medical Unit of Palo Alto VA Health Care System (PAVAHCS). C57/BL6, Ifnar1-/-, Casp1-/-, Il18-/-, Nlrp9b-/- were used as previously described (Lin et al., 2016 (link); Zhu et al., 2017 (link)). For all RV infections, sex and age-matched suckling pups at 5 days of age were used and whenever possible, littermates were used to minimize the effect of specific microbiome differences on the experiments.
IEC isolation by EDTA-DTT extraction method and flow cytometry of mature villous IECs were performed as previously described (Sen et al., 2012 (link)). Fecal shedding of viral antigens, and detection of RV replication by RT-qPCR were performed as described (Li et al., 2017 (link)).

OT-II+ TCRα−/− CD45.1+/+(OT-II) mice were kindly provided by Arlene Sharpe (Harvard Medical School, Boston, USA). Animals were housed in specific pathogen free and BSL2 conditions at The University of Chicago, and all experiments were performed in accordance with the US National Institutes of Health Guide for the Care and Use of Laboratory Animals and approved by The University of Chicago Institutional Animal Care and Use Committee.
