Dopamine-Loaded Transferrin-Conjugated Liposomes

Dopamine hydrochloride (DA∙HCl, MW = 189.64 g/mol), cholesterol (Chol), Triton X-100, N-(3-dimethylamino-propyl)-N’-ethylcarbodiimide hydrochloride (EDC), N-hydroxysulfosuccinimide (S-NHS) and Tranferrin (Tf) were bought from Sigma-Aldrich (Milan, Italy).
Hydrogenated soy phosphatidylcoline (Phospholipon 90H, PC) was a gift of Natterman Phospholipids GmbH (Koeln, Germany). 1,2-stearoyl-sn-glycero-3-phosphoethanolamine-N-[carboxy(poly(ethylene glycol)-2000)] (DSPE-PEG2000-COOH) was purchased from Avanti Polar Lipids (Alabaster, AL, USA).
For cellular transport studies luciferin yellow was bought from Sigma-Aldrich (Milan, Italy); Transwell® permeable supports were from Corning (Corning, NY, USA). All the media and supplements for cell culture were bought from Life Technologies, Thermo Fisher Scientific (Waltham, MA, USA). Other materials used in this study were of analytical grade.

Graphene Oxide Surface Modification

Aqueous slurry of GO nanosheets (3.73 mg mL−1) was provided by Graphenea (Donostia, Spain). The improved oxidation degree GO dispersion (1.5 mg mL−1) was supplied as well by Graphenea. Further information on the characteristics of the dispersions is available on the provider website. Chloroacetic acid (ClCH2COOH, ≥99.7%), hydrocloric acid (HCl, 37%), sodium hydroxide (NaOH, ≥98%), ethylenediamine (EDA, 99%), diethylenetriamine (DETA, 99%), triethylenetetramine (TETA, ≥97%), 1-Ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC, ≥98%) and N-hydroxysulfosuccinimide (S-NHS, ≥98%) reagent grade were purchased from Sigma Aldrich (Valencia, Spain) and used as received.

Dysprosium-Loaded Polymer Nanoparticles

Dysprosium nitrate hydrate (Dy(NO 3 ) 3 •5H 2 O, Sigma Aldrich, 99.99%), phosphoric acid (Sigma Aldrich, 85%), butanol Fig. 9 H&E staining of tissue sections of liver, kidney, spleen, and lung injected with saline (Ctrl) and with Dy57@PAA@PEG NPs.
(Sigma Aldrich, >99.5%), and 1-octanol (Merck, >99%) were used for synthesis. Poly(acrylic acid) (PAA, average M w ∼ 1800, Sigma-Aldrich) was used for NPs functionalization with PAA. For NPs functionalization with PEG, the following reagents were used as received: 1-(3-dimethylaminopropyl)-3ethyl carbodiimide (EDC -Sigma Aldrich, USA), N-hydroxysulfosuccinimide (s-NHS -Sigma Aldrich, USA) and α-methoxy-ω-amino PEG of 5000 Da (PEG, RappPolymer, Germany). Colloidal stability of NPs was evaluated using an aqueous solution of Phosphate Buffered Saline (PBS, Sigma Aldrich) that was prepared as follows: one tablet of PBS was dissolved in 200 mL water to obtain 137 mM NaCl, 2.7 mM KCl and 10 mM phosphate buffer, pH 7.4 at 25 °C. An aqueous solution of sodium tetraborate decahydrate (Sigma Aldrich, >99.5%) 10 mM was prepared and its pH was adjusted to 9 with NaOH. We call this solution borate buffer from now on.

Synthesis of Mono-Functionalized Heparin

Maleimide functionalized heparin was synthesized in-house following a previously established protocol (Tsurkan et al. 2013) where the molar ratio to heparin was reduced to achieve mono functionalization. Briefly, in an ice bath 100 mg of the heparin (Merck, Germany) were dissolved in 540 µl ultrapure water followed by the addition of 40 µl of 144 mg/ml 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and 80 µl of 41 mg/ml N-hydroxysulfosuccinimide (s-NHS) (Sigma-Aldrich, USA). The reaction was allowed to proceed for 20 min before adding 40 µl of 95 mg/ml of N-(2-aminoethyl) maleimide. The mixture was allowed to react overnight with continuous stirring at room temperature. Finally, the reaction mixture was dialyzed (molecular weight cutoff MWCO = 8 kDa) against 1 M NaCl for one day and then against deionized water for another day, concentrated, and lyophilized.
