Genetically Engineered Mouse Models

All animal husbandry and procedures followed the ethical guidelines outlined in the NIH Guide for the Care and Use of Laboratory Animals (https://grants.nih.gov/grants/olaw/guide-for-the-care-and-use-of-laboratory-animals.pdf), and all protocols were approved by the institutional animal care and use committee (IACUC) at Harvard Medical School. Slc6a2-p2a-Cre and Gfral-p2a-Cre mice were constructed as described below; Glp1r-ires-Cre (Williams et al., 2016 (link)), Calcr-ires-Cre (Pan et al., 2018 (link)), and Slc17a6-ires-Cre (Vong et al., 2011 (link)) mice were described before; mouse lines for cell-specific deletion and rescue of Glp1r were gifts from Randy Seeley (University of Michigan) (Sisley et al., 2014 (link)); Calca-gfp mice were from GENSAT; wild-type C57BL/6J (000664), Rosa26-LSL-L10GFP (024750), Chat-gfp (007902), Rosa26-LSL-tdTomato (007908), Gad2-ires-Cre (010802), and Rosa26-LSL-DTR (007900) mice were purchased (Jackson Laboratory). Single-cell sequencing was performed on male C57BL/6J mice; both male and female mice between 8-24 weeks old were used for all other studies, and no differences based on sex were observed.

Cre-Dependent Mouse Lines for Neuroscience Research

Crhr2-ires-Cre and Gabra1-ires-Cre mice were constructed as described below; Gpr65-ires-Cre, Glp1r-ires-Cre, Npy2r-ires-Cre, P2ry1-ires-Cre and P2×2/P2×3−/− mice were described before (Chang et al., 2015 (link); Finger et al., 2005 ; Williams et al., 2016 (link)); Vglut2-ires-Cre and loxP-L10-GFP mice were gifts from Bradford Lowell (Beth Israel Deaconess Medical Center); and wild type C57BL/6J (000664), Calb1-ires-Cre (028532), Chat-ires-Cre (006410), Chat-GFP (007902), Krt8-CreER (017947), Npy1r-Cre (030544), Piezo2-EGfp-ires-Cre (027719), Plcβ2−/− (018064), loxP-tdTomato (007908), loxP-ChR2 (012569), and loxP-DTR (007900) mice were purchased (Jackson Laboratory). The constitutive GCaMP3 allele (Rosa26-GCaMP3) was generated by breeding loxP-GCaMP3 (014538) with E2a-Cre mice (Jackson, 003314), and then crossing out the E2a-Cre allele. Single-cell sequencing was performed on male C57BL/6J mice; both male and female mice between 8–24 weeks old were used for all other studies, and no differences based on sex were observed.

Cre-Driver Mouse Lines for Neuronal Experiments

All animal procedures complied with institutional animal care and use committee guidelines. P2ry1-ires-Cre, Npy2r-ires-Cre, and Gpr65-ires-Cre were prepared using standard bacterial artificial chromosome (BAC) recombineering approaches (Figure S1), as previously described (Krashes et al., 2014 (link)) (see Extended Experimental Procedures). Chat-ires-Cre (006410), Chat-GFP (007902), lox-ChR2 (012569), and lox-tdTomato (007914) were purchased (Jackson). Vglut2-ires-Cre and lox-L10-GFP mice (Krashes et al., 2014 (link); Vong et al., 2011 (link)) were generous gifts from Bradford Lowell (Beth Israel Deaconess Medical Center). All Cre driver lines used are viable and fertile and abnormal phenotypes were not detected.
