Visualizing PCSK9 Expression in oxLDL-Treated Cells

Following stimulation with various concentrations of oxLDL (50 and 100 µg/ml) for 24 h, the cells were fixed with 4% formaldehyde for 30 min. Following incubation with 0.1% Triton X-100 at room temperature for 30 min, the cells were blocked with goat serum for 1 h at room temperature. After being washed thrice with PBS, the cells were incubated with diluted PCSK9 antibody (1:100; #85813, Cell Signaling Technology, Inc.) at 4°C overnight. After being washed 5 times with PBS, the cells were incubated with Alexa Fluor-conjugated secondary antibody (1:50, A32731, Invitrogen; Thermo Fisher Scientific, Inc.) in the dark for 1 h at room temperature. Finally, the nuclei were stained with 4′,6-diamidino-2-phenylindole (Beyotime Institute of Biotechnology, Inc.) for 5 min at room temperature. After being washed 5 times with PBS, the cells were observed under a fluorescence microscope (IX3; Olympus Corporation).
