Histological Evaluation of Lymphoid Tissues

Peripheral LN, splenic, and thymic tissue were evaluated by histology. Standard H&E staining was performed on all tissue. On LN tissue, immunofluoresence (IF) using anti-CD3 and anti-CD20 (Dako, Carpinteria, CA) staining was performed in order to evaluate germinal center formation. We also examined the thymus for evidence of binding of RATG using a biotinylated goat-anti-rabbit polyclonal antibody (BA-1000-1.5; Vector Laboratories, Burlingame, CA) to detect RATG. A primary mouse anti-cytokeratin Ab (MS-611-P0; Neomarkers, Fremont, CA) followed by a secondary biotinylated horse anti-mouse Ab (BA200, Vector Laboratories, Burlingame, CA) as a positive control. Samples were developed using the Vector Elite Kit (PK-6100; Vector Laboratories, Burlingame, CA).

Multimarker Immunohistochemistry of Murine Tissues

Sections were then incubated sequentially with the primary antibody overnight and followed by 1 h incubation with biotinylated anti-mouse (Vector Labs; BA-9200-1.5) or rabbit (Vector Labs; BA-1000-1.5) secondary antibodies in a 1:500 dilution. The Streptavidin-HRP and DAB detection kit (Vector Labs) were used according to the manufacturer’s instructions. For quantification, positive cells were counted from at least three random slides of each mouse for a total of 4–8 pairs.

Quantifying Phosphorylated CREB in Rat Brains

Rat brains were prepared after conditioning for immunohistochemistry to analyze phosphorylated CREB (pCREB)–positive cells. Brains were sectioned and treated with rabbit anti-pCREB serum (1:1000; EMD Millipore 06-519) and biotinylated goat antirabbit IgG antibody (1:200, Vector Labs, BA-1000-1.5), and revealed by ABC kit (1:50, Vector Labs, PK-6100) and DAB-Ni solution (Vector Labs, SK-4100). Images were captured using a Nikon microscope and analyzed with QuPath software (45 ; https://qupath.github.io) for automatic cell detection and quantification. pCREB-positive cells in specified brain regions were counted, and densities were calculated.
