Cell Culture Protocol for Various Cell Lines

L-929 cells (ATCC) were purchased from the Chinese Academy of Sciences (Shanghai, China) and cultured in Dulbecco’s modified Eagle’s medium (DMEM) containing 10% fetal bovine serum (FBS) (Gibco, Grand Island, NY, USA) and 1% penicillin/streptomycin (100 units/mL) in a humidified atmosphere at 37 °C with 5% CO2.

Culturing MDA-MB-468 and 3T3 Cells

Fibroblast Culturing and Rat Housing

Establishing Mouse and Rat Cell Lines

Isolation and Culture of Immune Cells

Mouse splenic T cells were obtained from BALB/c, C57BL/6, OT-I, or OT-II mice via purification over nylon wool.

MCMV Infection and Antibody Response

B6 TECs, BALB/c TECs or BALB/c 3T3 cells (American Type Culture Collection, Manassas VA) were infected with MCMV-GFP virus at a multiplicity of infection (MOI) of 1 for each cell type. At 72 hours post infection, cells were detached and incubated with either MCMV− or MCMV+ B6 sera, or with transplant sera (D−/R− or D+/R+) at 1:20 dilution for 1 hour on ice. MCMV uninfected cells were also incubated with sera. Cells were washed and stained for viability (Ghost Dye UV450, TONBO Biosciences, San Diego CA), EpCAM-PE (CD326, Clone G8.8, Biolegend, San Diego CA), and anti-mouse IgG1-APC (Clone A85–1, BD Biosciences, San Jose CA). Samples were analyzed using a LSRFortessa (BD Biosciences) and FlowJo v10 software (TreeStar Inc, Ashland OR). Experiments were performed 3 times independently.

FITC-conjugated goat anti-mouse immunoglobulin G Fcγ antibody, horseradish peroxidase (HRP)-conjugated streptavidin, and HRP-conjugated goat anti-mouse IgG Fcγ antibody were purchased from Jackson Immuno Research Labs (West Grove, PA, U.S.A.).
BALB/c 3T3 cells (American Type Culture Collection, Manassas, VA, U.S.A.) were maintained at 37 °C with 5% CO2 and grown in culture medium containing DMEM, 10% BCS, and 100 units/mL of penicillin and streptomycin (Invitrogen, Calsbad, CA, U.S.A.). Expi293 cells were maintained in Expi293 Expression Medium (Invitrogen, Calsbad, CA, U.S.A.) at 37 °C and 120 rpm.
